Rationales: Aging is the process of degenerative change in the tissues and organ function.With the development of society and economy,life expectancy has increased,but the problems of aging population and health and diseases of the elderly population are more serious.The study of anti-aging drugs is a subject what the researchers around the world study on.Guilingji,a famous traditional Chinese medicine prescription,is an existing compound preparation that was proved by more than 400 years of clinical experiment.28 traditional Chinese medicines,including velvet antler,red ginseng,hippocampus,beetle,eucommia,shiyan,asparagus,and daqing salt,are involved in the prescription.Moreover,Guilingji is also a famous anti-aging prescription,which has the effect of delaying aging.However,the research of anti-aging of Guilingji is less and the mechanisms of it is unclear.In recent years,drosophila is a typically model organism for anti-aging drug research.Therefore,the model of drosophila was used to study the effect of lifespan and the activity and mechanism of anti-aging after Guilingji treatment.Objective: In this study,the anti-aging activity of Guilingji was verified and the mechanism of delaying age was explained,which provided a basis for the subsequently deeper research.Methods: 1.The effect on lifespan of drosophila after Guilingji treatment..The model of drosophila was used for lifespan test.The drosophila was treated with different concentrations(0,0.5,1,2,4and 8mg/m L),different dosing periods(Youth: 1-day-old drosophila;elderly:30-day-old drosophila and 45-day-old drosophila),and different modes of administration((1)30-day-old drosophila were treated with Guilingji:(A)continuously treated with Guilingji to 45-day-old,(B)until the death with once every other day treatment;(2)45-day-old drosophila were treated with Guilingji:(C)until the death with continuous treatment.2.The effect and mechanism of Guilingji on the behavioral ability of natural aging drosophila.Drosophila were treated with Guilingji according to the period,dose and manner obtained in the above experiment.Sexual activity,exercise activity,anti-stress ability(cold,hot,hunger)were applied to evaluate activity of Guilingji;the contents of catalase(CAT)and superoxide dismutase(SOD)were measured to evaluate the antioxidant activity of Guilingji;UPLC-MS/MS based metabonomics was employed to reveal the anti-aging mechanisms of Guilingji in drosophila.Results: 1.The results of drosophila treated with drug at young showed that compared with control group,the survival rate,average life expectancy and maximum lifespan were lower in Guilingji-treated group,suggesting that Guilingji has no effect on improving lifespan.And the optimal dose for feeding 30-day-old drosophila is 4 mg/m L.Both A and C administration methods can improve the lifespan of drosophila,and significantly increase the average lifespan,median survival time and maximum lifespan of drosophila.Thus,the Guilingji can prolong the lifespan of the old drosophila and has an anti-aging effect.2.The Guilingji has significantly improved the active ability of drosophila,in other words,the Guilingji has significantly improved the ability of climbing of the drosophila.And then the health condition of it was improved,and the quality of life was increased.Unfortunately,the ability of reproductive and anti-stress of Guilingji-treated drosophila was not observed.3.Guilingji didn’t significantly increase the activity of CAT and SOD in drosophila.15 aging-related differential metabolites,including L-glutamine,pyroglutamic acid,choline,L-palmitoyl carnitine,L-acetylcarnitine,adenosine 5’-monophosphate,L-glutamic acid,linoleoyl carnitine,proline,lysophosphatidylethanolamine Lyso PE(16:1),lysophosphatidylcholine Lyso PC(18: 3),Lyso PC(16:1),Lyso PC(16:0),adenosine and Elaidic carnitine were found after analyzing the metabolic profiles of different groups of drosophila.Amino acid metabolism pathways were major pathway,including glutathione metabolism,D-glutamine and D-glutamate metabolism,arginine and proline metabolism,alanine,aspartame Acid and glutamate metabolism,which the 15 potential aging biomarkers participated in.The results showed that Guilingji has the effect of delaying aging by regulating the disorder of amino acid metabolism during aging.Conclusion: Guilingji can significantly prolong the lifespan of old-aged drosophila,improve exercise vitality,and improve the health conditions of drosophila,which are closely related to improve metabolic disorders by regulating amino acid metabolism pathways.However,Guilingji is not suitable for young drosophila.The effects on drosophila sexual activity and anti-stress need to be adjusted for further study.The activity of antioxidant enzymes is not positively correlated with lifespan.
