| Moderate UV exposure is inevitable during normal life activities and does not have a major impact on health.However,overexposure to UV,particularly UVB radiation,leads to excessive ROS and inflammatory cytokine,putting overexpression of matrix metalloproteinase(MMPs),which leads to the degradation of extracellular matrix and the appearance of roughness,coarse wrinkles,erythema,and dandruff.As a result of these adverse health effects,researchers are exploring the potential of using antioxidants and anti-inflammatory substances to increase the skin’s resistance to UVB irradiation.Recently,much effort has been directed towards the development of effective natural compounds for cutaneous photoprotection.Snake oil which is a traditional natural skin care product,has been proved to have good anti-inflammatory,antioxidant.However,little is known about the biological effects of snake oil such as photoprotection,and currently the products of snake oil on the market are relatively single(cream).Based on mentioned above,In the present study,we will explore whether snake oil could prevent light damage by building an animal model of light damage,and develop new dosage forms of snake oil gel products with beeswax and monoglyceride as gel factors.In this paper,main conclusions are as follows:1.Topical application of snake oil and its fatty acids can markedly improve the skin macroscopic surface and epidermal thickening of mice induced by UVB irradiation.Histological observation demonstrated that snake oil and its fatty acids could evidently decrease the degradation of collagen fibers and elastic fibers.Meanwhile,snake oil and its main fatty acids could significantly inhibit the decrease of antioxidant enzymes and the increase of MDA,and it’s not significant different between fatty acids and snake oil.Analysis of inflammatory factors and inflammatory cells showed obvious reduced,and the effect of snake oil is markedly better than its main fatty acids.In addition,snake oil and its fatty acids pretreatment evidently inhibited the abnormal expression of MMP-1.Based on the results of various tests and the exploration of the current mechanism of light damage,we believe that snake oil and its main fatty acids conspicuously counteracted the UVB-induced increase of MMP-1 secretion through inhibiting the decrease of antioxidant enzymes(SOD,CAT and GSH-Px)activity and the increase of inflammatory factors,then reversed the UVB-mediated collagen and elastic fibres destruction,and thereby protected skin from photodamage.Furthermore,we found that the main fatty acids played a major role in the antioxidant activity of the snake oil,followed by anti-inflammatory effects by comparing the effects of snake oil and its main fatty acids in various indicators.2.In this study,the effect of optimum ratio of beeswax and monoglyceride in the preparation of snake oil-based gel oil were studied.Results showed that the presence of a small amount of monoglyceride(beeswax/monoglyceride=8:2)could promote the generation of gel-like dendritic crystals,increase the number of crystals of α,β and β’,but reduce thixotropy and hardness.As the content of monoglyceride increases(0:10<beeswax/monoglyceride<8:2),the elongated dendritic crystals gradually shorten to disappear,and the synetic crystals gradually disappear,but the thixotropy is increased.The analysis of the properties of snake(deinagkistrodon acutus)oil organogel by the addition ratio of beeswax and monoglyceride was carried out to determine the optimal ratio when beeswax/monoglyceride=4:6.At this time,oil-binding capacity of snake oil organogel was 1.8%,the hardness was 242 g,and the viscosity recovery value was 37%.The snake oil gel product with high snake oil content was developed based on this result.In addition,low snake oil skin balm was developed. |
