Objective: This subject used obstructive jaundice model mice as experimental subjects,with silybin lavage way of intervention to investigate the mechanism of silybin on the repair and regeneration of hepatocytes in obstructive jaundice mice by iNOS/NF-κB signaling pathway.Methods: 30 KM mice were randomly divided into twenty mice with obstructive jaundice were constructed and randomly divided into model group(M)and drug group(S),and 10 patients with blank group(SO).In group S,the daily dose of human and the body weight coefficient of the mice were combined with silybin 30 mg/kg.d for a total of 7 weeks.At the end of the drug intervention program,the materials were taken.The mice were fasted and banned on the night before.The next day,the mice were anesthetized.The mice were anesthetized after complete anesthesia,the surrounding tissues were freed,and the liver was fully exposed.The liver was taken out,and the specimens were sliced and treated with hematoxylin-eosin staining.The pathological changes of liver tissue were observed under microscope.mouse liver tissue.General behavioral,morphological observation,Elisa enzyme-linked immunosorbent assay,liver tissue protein assay and qRT-PCR were used to study the improvement of silybin on the liver tissue of obstructive jaundice mice to investigate the mechanism of silybin on the repair and regeneration of hepatocytes in obstructive jaundice mice by iNOS/NF-κB signaling pathway.Results: M group mice had severe yellow skin staining,dark urine,light gray stool,and obvious relief in group S.HE staining showed that the hepatocytes in the SO group were arranged neatly,The hepatic lobule is complete,no deformation and necrosis.and the hepatic cords in the M group were disordered,and a large number of vacuoles and nuclear fission appeared.Liver cells atrophy,fracture,nucleus pycnosis,etc,Visible mass accumulation of Inflammatory cells;Compared with the M group,the hepatocytes in the S group were arranged more regularly,Local degeneration of fat cells,and the area of flaky necrosis was significantly reduced.serological results:Compared with group M,the levels of total bilirubin(TBIL),alanine aminotransferase(ALT)and aspartate aminotransferase(ALT)in group S decreased significantly;iNOS,NF-κB,TNF-α,Par-4 protein in liver tissue The content is significantly reduced.The results of qRT-PCR showed that the expression levels of iNOS,NF-κB,TNF-α and Par-4 mRNA in M group were significantly higher than those in S group.Conclusions: Silibinin has an intervention effect on obstructive jaundice in mice,which can enhance the immune function of mice with liver injury induced by obstructive jaundice,thus significantly reducing the degree of liver damage induced by blocking yellow.The mechanism may be to alleviate the development of liver injury by regulating the expression of cytokines related to the iNOS/NF-κB pathway and maintaining the integrity of hepatocytes,Enhance the repair capacity of obstructive jaundice liver cells in mice.
