The acute toxicity of α-amanitin extracted from fruit body of Amanita exitialis Z. L. Yang&T.H. Li was studied. The protective effects of Ganoderma lucidum extract, silybin, vitamin C and Legalon on a-amanitin-induced liver injury were compared, and the protective effect of Legalon on liver injury was analyzed systematically on the dosage and time. The results are as follows:1. The semi-lethal dose(LD50) of α-amanitin in mice is0.4130mg/kg,and the95%confidence limits span from0.3563mg/kg to0.4789mg/kg, the results were consistent exactly with the data reported by various literature. After toxin injection, the action of mice is slow, the severe cases even dead, anatomical observation found that liver became pale colored, even with white spots, indicating that poisoning can cause liver cell necrosis.2. The protective role of Ganoderma lucidum extract, silybin, vitamin C and Legalon on the amatoxin-induced liver damage were compared. The results showed that various biochemical indicators of amatoxin poisoning group were significant different from the normal control group. The best of the treatment group was Legalon and showing a concentration-related, followed by the the silybin group, the vitamin C group has no treatment effect.3. The hepatoprotect of different doses Legalon for hepatic injury due to α-amanitin were analyzed. the results indicated that the protective effect was dose-related, the doses increaser(20to500mg/kg/day), the better it reduce the role of the liver transaminase activity. When doses up to200mg/kg/day and500mg/kg/day (concentration near the solubility limit), transaminase activity reached the lowest value.4. The injection time of Legalon administration for the α-amanitin-induced liver injury were studied. the results showed that the earier injection the less liver injury, especially in advance administration Oh biochemical indicators close to the normal control group, indicating that prevention obvious, but in reality this is basically impossible. The results tell us that as soon as possible treatment after the mushroom poisoning is necessary.5. The mice mortality of Legalon administration and the control were statisticed. After120h of administration, no death in the normal group, the mortality of amanitin poisoning group without Legalon treatment attained to94%, while the mortality rate of Legalon group was only55%, it indicated that the Legalon has significant treatment effect.6. The experimental results showed that amanita poisoning is related to its oxidation, the four drugs can increase the liver tissue superoxide dismutase (SOD), the activity of catalase (CAT) and reduce malondialdehyde (MDA) capabilities, the Legalon achieved to extremely significant effect.
