Objective: To analyze the relationship between circulating tumor cell count and clinical data such as tumor TNM staging,and to explore the clinical application value of circulating tumor cell detection as an auxiliary examination method for colorectal cancer patients.Methods: 1.Peripheral blood was collected from 56 patients with newly diagnosed colorectal cancer,and the age,sex,postoperative pathology,Carcinoembryonic antigen(CEA),and Carbohydrate antigen 19-9(CA199),were collected.),Carbohydrate antigen,(CA242),Cluster of differentiation 4(CD4),Cluster of differentiation 8(CD8)and other clinical data.Another 20 healthy volunteers and colorectal benign tumor patients were used as the control group.The above samples were detected by immunomagnetic beads sorting combined with fluorescent staining to detect circulating tumor cells.2.Compare the CTCs count with the clinical features of patients with colorectal cancer,and analyze the differences between the groups.3.The receiver operating characteristic curve(ROC)was drawn,and the logistic regression was introduced into the ROC curve to evaluate the application value of joint detection.Results: 1.Patients with different TNM stage,tumor diameter,CEA level and CA242 level had different expression levels of circulating tumor cells,and the difference was statistically significant(P=0.000,P=0.022,P=0.011,P=0.014).2.Patients with different tumor differentiation and CD8 expression levels had different levels of CEA expression,and the difference was statistically significant(P=0.014,P=0.016).The expression levels of CA199 were different in patients with different tumor diameter and CD8+ expression levels,and the difference was statistically significant(P=0.017,P=0.009).3.The area under the ROC curve of 3CTCs,CEA,CA199,and CA242 were 0.861,0.841,0.584,and 0.760,respectively.The area underthe ROC curve of triple and quadruple after logistic regression was 0.884 and 0.946,respectively(P<0.05).Conclusion: The circulating tumor cells of patients with newly diagnosed colorectal cancer have higher expression levels in patients with late TNM stage,long tumor diameter,high CEA and high CA242 than those with early TNM stage,short tumor diameter,low CEA and low CA242.Circulating tumor cell detection can be combined with traditional tumor markers and supplemented by the current colorectal cancer auxiliary examination system,which has clinical application value.
