Phellinus linteus polysaccharide(PLP)is the main biological active ingredient in Phellinus linteusis.It has many biological activities such as anti-tumor,immune regulation,anti-oxidation and hypoglycemic.The anti-inflammatory and lipid-lowering effects of Phellinus linteus polysaccharide are still in their infancy.In this paper,Phellinus linteus polysaccharide was used as the research object,in vitro inflammatory model of ICR mice induced by dextran sulfate sodium(DSS)and lipopolysaccharide(LPS)-induced RAW 264.7 in vitro inflammation model,and high fat diet(HFD)to induce the hyperlipidemia model in C57BL/6 mice and the oleic acid(OA)-induced HepG2 hepatoma cell fat accumulation in vitro model to evaluate the anti-inflammatory and lipid-lowering functions of Phellinus linteus polysaccharide and its molecular mechanism.To provide a theoretical basis for the development of Phellinus linteus polysaccharide as a functional food for slowing colitis and hyperlipidemia.Drug development for mitogen-activated protein kinases(MAPK),peroxisome proliferator-activated receptors(PPARs),and AMP-activated protein kinases(AMPK)for colitis and hyperlipidemia provide a theoretical basis for the prevention or adjuvant treatment of colitis and hyperlipidemia.1.Anti-inflammatory effect of PLP and its molecular mechanismSixty mice were randomly divided into 3 groups,and DSS group and DSS +PLP group mice received 2.0%DSS and PLP was orally administered at 500 mg/kg/day.Our data showed that PLP administration obviously improved the health status of mice and inhibited DSS-induced pathological alterations and significantly reduced inflammatory cytokine expressions in the colonic tissues.In lipopolysaccharide-induced inflammation cell model,PLP supplement also significantly reduced inflammatory cytokine expressions and inhibited MAPK pathway as well as the translocations of NF-κB and AP-1.Meanwhile,PLP supplement regulated PPARa and PPARy phosphorylation and blocked MAPK activation.Taken together,PLP exhibits anti-inflammatory function and its molecular mechanism may be involved in MAPK and PPAR signal pathways,which reduce the expressions of inflammatory cytokines.2.Lipid-lowering effect of PLP and its molecular mechanismSixty mice were randomly divided into control group,high-fat diet group and Phellinus linteus polysaccharide-protected group,the 500 mg/kg.bw/day PLP was added by intragastric administration.The results showed that the body weight,liver weight and adipose tissue weight of the mice were significantly decreased after the protection of the PLP,and the contents of total cholesterol(TC),triglyceride(TG)and low density lipoprotein cholesterol(LDL-C)were decreased.Inhibits the reduction of high density lipoprotein cholesterol(HDL-C).Phellinus linteus polysaccharide can reduce the size of lipid droplets and the size of fat cells in liver tissue.Western blotting and RT-qPCR confirmed that the protection of Phellinus linteus polysaccharide could significantly regulate the expression of lipid metabolism-related genes in high-fat animal model and oleic acid-induced hepatic cell lipid accumulation cell model.Phellinus linteus polysaccharide also inhibits the expression of inflammatory factors.In conclusion,our data indicate that Phellinus linteus polysaccharide treatment can regulate lipid metabolism-related gene expression and inhibit lipid accumulation in liver cancer cells induced by high fat diet in mice obese and oleic acid.
