Purpose:To detect the expression levels of human transmembrane protein 106b(TMEM106b)in the different grades of meningiomas and the normal meninge tissue,and to evaluate the Relevance and the TMEM106 b expression levels in the different grades of meningiomas and the normal meninge tissue.To further explore and detect the occurrence and progression mechanism of meningioma,So as to provide basis research in the further in the way of development and treatment about meningioma.Method:Sixty patients with meningioma from the Department of Pathology,the First Affiliated Hospital of Nanchang University from January 1,2017 to January 1,2019,and 20 normal meninge tissue from patients with craniocerebral trauma were selected as controls.The TMEM106 b expression levels in the different grades of meningioma and normal meningeal tissues was detected by Q-PCR.The TMEM106 b proteins in the different grades of meningiomas and normal meninge tissues were detected by Western blot.The expression of TMEM106 b protein in the meningioma and normal meninge tissues,as well as different expression levels of meningioma tissues,were analyzed.The analysis results were statistically significant(p<0.05).Result:1.Q-PCR method is used to detect the TMEM106 b mRNA expression levels in the normal meninges is low,and set the TMEM106 b mRNA expression levels in normal meninges as the control group,the TMEM106 b mRNA expression levels in the grade of WHO I meningioma was 1.88±0.16,the TMEM106 b mRNA expression levels in the grade of WHOⅡmeningioma was 2.91±0.28,the TMEM106 b mRNA expression levels in the grade of WHO Ⅲ meningioma was 1.57±0.28.After statistical analysis,the expression levels of TMEM106 b between normal meninges and meningiomas was significantly different(p<0.05).There was no correlation between the expression levels of TMEM106 b in the different grades of meningiomas(p>0.05).2.Western blot was used to determine the low expression levels of TMEM106 b protein in normal meninges,and the low expression levels of TMEM106 b protein in normal meninges was set as the control.The expression levels of TMEM106 b in grade I meningioma was 1.91±0.18 by Western blotting,the expression levels of TMEM106 b in grade II meningioma was 3.04±0.41,and the expression levels of TMEM106 b in grade III meningioma was 1.62±0.38.The TMEM106 b expression levels was not statistically significant between the different grades of meningiomas(p>0.05).Conclusion:(1)The TMEM106 b expression levels in the meningiomas was higher,but it is low expression levels in normal meninges in this experiment,indicating that the expression levels of TMEM106 b may be related to the meningioma.It is may related to the occurrence and development of the meningioma.(2)The TMEM106 b expression levels was not correlated with the different grades of meningiomas.However,the TMEM106 b expression levels in meningioma was high,the higher expression levels in the meningioma was not increase by proportional with the different grades of meningiomas It indicated that the TMEM106 b expression levels was not significantly correlated with the different grades of meningiomas.And it may not be one of the indexes to infer the classification of meningioma.
