The Expression And Significance Of HER-2 And TMEM106B In The Process Of Cell Proliforation And Angiogenesis Of Meningioma | | Posted on:2018-04-12 | Degree:Master | Type:Thesis | | Country:China | Candidate:Z H Ding | Full Text:PDF | | GTID:2334330518962412 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | ObjectiveThis study is just to detect the expression rate of the proteins of HER-2,TMEM106 B,VEGF and CD34,and discuss the relationship between HER-2and TMEM106 B with meningioma cells proliferation and angiogensis,therefore,in order to further understand the mechanism of the occurrence and progress of meningiomas,which would provide experimental data for the following targeted therapy of meningiomas.Methods:Selection on January 1,2007-December 31,2015 in the pathology department of the First affiliated hospital of NanChang University paraffin specimens of 50 cases postoperatively in patients with meningioma,and 20 cases of normal meninges paraffin tissue as control meningeal tissues are all from removal of intracranial hematoma after craniocerebral injury,and add to patients with bone decompression.the immunohistochemical method of MaxVisionTM was taken to respectively detect the proteins of HER-2,TMEM106 B,VEGF and CD34 in the two groups of samples,and use the UTHSCSA Image Tool graphic analysis software to calculate the microvascular density(MVD).Results:(1)HER-2 in this group of meningioma tissue have higher postive detection rate36%(18/50),but the expression of HER-2 protein in normal meninges tissues are negative,the expression of both have statistically significant differences(X2=9.692,P<0.05).But the further study shows that HER-2 positive rate is not increased with the degree of malignant meningioma tissue,the protein of HER-2 positive rate in the WHOI meningioma tissues is 28.6%(6/26),HER-2 positive rate in the WHOII meningioma tissues is 44.4%(8/18),WHOIII level positive rate of meningioma tissueare 36.4%(4/11),the difference was not statistically significance(X2=2.007,P>0.05).(2)TMEM106B in this group of meningioma tissue have higher postive detection rate82%(41/50),but the expression of TMEM106 B protein in normal meninges tissues are negative,the expression of both have statistically significant differences(X2=39.586,P<0.05).But the further study shows that TMEM106 B positive rate is not increased with the degree of malignant meningioma tissue,the protein of TMEM106 B positive rate in the WHOI meningioma tissues is 81%(17/21),the proteins of TMEM106 B positive rate in the WHOII meningioma tissues is77.8%(14/18),WHOIII level positive rate of meningioma tissue are 90.9%(10/11),the difference was not statistically significance(X2=0.004,P>0.05).(3)The expression rate of MVD and VEGF in normal meningioma tissues are negative,and the expression rate of MVD and VEGF in meningioma tissue was significantly increased by(18.04 ± 19.85)and 70%(35/50),the differences were statistically significant(X2=9.692 P<0.05,T=9.427 P<0.05),the levels in meningioma are as follow:the level of CD34 and VEGF in WHOI meningioma tissues are(5.77±10.18)and 66.7%(14/21),the level of CD34 and VEGF in WHOII as well as WHOIII meningioma tissues are(26.92±20.53)and 72.41%(21/29).the differences were statistically significant.(4)According to the Spearman rank correlation analysis,HER-2 and VEGF were positively correlated,HER-2 and CD34 were also positively correlated.Positive correlation between HER-2 and VEGF(r=0.113,P<0.05),and MVD were positively correlated(r=0.058,P<0.05).TMEM106 B protein was positively correlated with VEGF(r=0.293,P=0.039)and was positively correlated with MVD(r=0.146,P<0.05);VEGF and MVD were positively correlated(P<0.05,r=0.017).HER-2 and TMEM106 B were positively correlated(P<0.05,r=0.115).Conclusion:(1)HER-2 and TMEM106 B in meningioma cells there were over expression,but no significant degree of malignancy and the positve expression rate of meningioma both HER-2 and TMEM106 B are involved in the angiogenesis and proliferation of meningioma cells.(2)the protein of HER-2 may lay a influence on by adjusting TMEM106 B meningeal cells proliferation and angiogenesis. | | Keywords/Search Tags: | TMEM106B, HER-2, VEGF, CD34, MVD, Immunochemistry | PDF Full Text Request | Related items |
