Objective Recently,substantial studies from domestic and overseas have proved that the gut microbiota is inextricably linked to the diseases of human body’s digestive,immune,metabolic,nervous and neuropsychiatric systems and revealed the indispensable role of the gut microbiota in human body.This study that examined the gut microbiota’s structure of HT patients and compared them with healthy people,aims to find out the specific changes of the gut microbiota’ structure in patients with hyperthyroidism,and explore the role of these microbial communities in the etiopathology and development of hyperthyroidism.Methods Fifteen HT patients and 15 healthy volunteers were recruited to establish HT group and HC group,respectively.The 30 fecal samples from HT group and HC group were collected.Microbiological analysis of the gut microbiota in the 30 fecal samples were performed by 16 S r RNA sequencing technology,and the results of the two populations were compared on the Phylum and Genus level of the gut microbiota.On the basis of comparison,the difference of the richness,diversity and structure composition between the two group were found.The correlation between serological and functional indicators related to hyperthyroidism and the gut microbiota with different abundance between the two groups was analyzed.Predictive analysis of the function of the gut microbiota with different abundance between the two groups in hyperthyroidism was performed by the KEGG database.Results At the Phylum level,the richness indexes of the gut microbiota in HT group were lower than HC group,while some diversity indexes in HT group were higher than HC group.The Venn diagram showed that there were 13 phyla in HT and HC group,all of which were dominated by Bacteroidetes,Proteobacteria,Actinobacteria and Firmicutes.In the composition of the gut microbiota,the relative abundance of Bacteroidetes and Proteobacteria in HT group were more than HC group,while the relative abundance of Actinobacteria and Firmicutes were less than HC group.All the differences were not statistically significant(P>0.05).At the Genus level,the richness,some diversity indexes(sobs,ace,chao,shannon)and coverage index of the gut microbiota in HT group were lower than HC group,while simpson index of HT group was higher than HC group(P<0.05).The Venn diagram showed that 246 genera were detected in HT and HC group.HC group contained 51 species of endemic bacteria,while the bacterial taxa of HT group had a significant decrease,with only 7 genera.The relative abundance of Eubacterium rectale group,Bifidobacterium,Eubacterium hallii group,Prevotella 9 and Fusicatenibacter in HT group were less than HC group,while the relative abundance of Bacteroides and Escherichia Shigella in HT group were more than HC group.The difference of the relative abundance of Eubacterium rectale group between the two groups were statistically significant(P<0.05).And the AUC of ROC curve was 0.72.What’s more,in the correlation analysis of immune function-related metabolism,it was found that Eubacterium rectale group was negatively associated with the metabolic pathway of Bacterial invasion of epithelial cells(r=-0.367,P=0.046).Conclusion 1.Ecological disorders of the gut microbiota exists in hyperthyroidism patients.2.The relative abundance of Eubacterium rectale group may be able to distinguish hyperthyroidism patients from healthy people.3.Eubacterium rectale group is associated with the metabolic pathway of Bacterial invasion of epithelial cells.4.Eubacterium rectale group may provide a new perspective for the diagnosis and therapy of hyperthyroidism. |
