Objective:To analyze the quantitative differences of target bacteria in intestinal microflra of NGT people and T2DM people in Uygur and Kazak population. To analysis the structure characteristics of the intestinal microflora of NGT people and T2DM people in Uygur and Kazak, respectively, to explore the correlation between intestinal microflora and T2DM. Methods:(1)Relative quantification of 16SrDNA Real Time PCR was used. (2) We used 16SrDNA-PCR-DGGE to analyze the different structure of intestinal microflora in NGT people and T2DM people. The cloning and sequencing of the different bands were to define bacterial species by sequence alignment in Genebank database.Result:(1)Compared the initial copy number of target bacteria in intestinal microflora between NGT group and T2DM group of the two national. The number had significant differences (P<0.05). (2) Bilophila wadsworthia and Thermus thermophilus was the dominant bacterial of NGT group in Uygur; Stenotrophomona, Cellvibrio sp, Thermus, Zymomonas was the dominant bacterial of T2DM group in Uygur; Crossotus was the dominant bacterial of NGT group in Kazak; Thermus, Eubacterium and Fusobacterium was the dominant bacterial of T2DM group in Kazak. Conclusion:The changing of target bacteria may be related to the onset of Type 2 diabetes mellitus in Uygur and Kazak. Based on the technical 16SrDNA-PCR-DGGE to analyze the genetic fingerpringting, we had a preliminary understanding of intestinal microflra... |
