Regulatory Effect Of SOCS3 On The Expression Of Indoleamine 2,3-dioxygenase At The Maternal-fetal Interface Of Early Pregnancy | | Posted on:2020-09-27 | Degree:Master | Type:Thesis | | Country:China | Candidate:S X Li | Full Text:PDF | | GTID:2404330575976523 | Subject:Obstetrics and Gynecology | | Abstract/Summary: | PDF Full Text Request | | Objective:The reason why embryos are not rejected is immune tolerance that depends on the maternal immune system during pregnancy,and the maternal-fetal interface is an important part of the regulatory mechanism of immune tolerance.In this study,the expression and correlation of IDO and SOCS3 were explored by studying the expression and correlation of the two proteins under the action of pregnancy-like estrogen and its inhibitor fulvestant,the regulation of the immune tolerance mechanism of embryos to the maternal-fetal interface was further explored.Methods:Thirty-five normal pregnant women investigated in the study were selected from those who under legal termination at the Affiliated Hospital of Guizhou Medical University,collection of their villi and decidua tissues between October 2016 to October 2018.The age of the women was 28.40±4.52 years;the gestational age of the subjects was 58.45±6.30 days.Blocks of chorionic villi and the decidua were cultured 48 h in media containing varying concentrations of 17-estradiol and estriol respectively.at the same time increseing estrogen inhibitor fulvestant to the blocks of chorionic villi and the decidua were cultured 48 h.Western blot and qPCR analysis were used to detect the expression of indoleamine 2,3-dioxygenase and suppressors of cytokine signalling 3 in chorionic villi and the decidua in cultured tissuesand.RESULTS: Both indoleamine 2,3-dioxygenase and suppressors of cytokine signalling 3 expressed in chorionic villi and decidua.The expression of IDO was upregulated and SOCS3 was downregulated the most,in blocks of chorionic villi and the decidua that were cultured in medium containing 17-estradiol at a concentration of 10ng/ml or estriol at a concentration of 1ug/ml,respectively.And estrogen inhibitor fulvestant made The expression of IDO was downregulated and SOCS3 was upregulated.CONCLUSIONS:At a concentration similar to that present during pregnancy,estrogen hormone and its isomer may upregulate the expression of indoleamine 2,3-dioxygenase via downregulated SOCS3,which implies that estrogen prevents allogeneic fetal rejection and may be used as an immunomodulator. | | Keywords/Search Tags: | estrogen, fulvestant, chorionic villi, decidua, indoleamine 2,3-dioxygenase, suppressors of cytokine signalling 3, maternal–fetal tolerance | PDF Full Text Request | Related items |
