Purpose:There are many studies on changes after chronic injury,but little is known about the early response of tendon to damage caused by load accumulation,so the relationship between the stages of inflammatory response in the early stage of tendinopathy is not clear.Therefore,on the quantitative jumping animal model successfully established in the laboratory,I studied the changes of the main inflammatory factors IL-1β,TGF-β1,IL-6 and p38 MAPK in the acute phase after high-intensity exercise,and explored the acute changes under the high-intensity load.Methods:34 18-week-old female New Zealand white rabbits weighing 2.5-3.0 kg were randomly divided into control group(n=4);jumping group 1 day(n=5),3 days(n=5),5 days group(n =5);Jump + p38 inhibition group 1 day,3 days,5 days groups(JUMP-p381 D,3D,5D,each n = 5),SB203580(0.5 mg / kg body weight)was intraperitoneally injected after each training.The experimental group performed electrical stimulation jump training according to the training program,the control group did not train,and the same pre-adaptation process was completed,and the feeding time was the same.Results:Histological changes HE staining: In the CON group,the tendon fibers were arranged in a wavy shape,the cells were relatively uniform,and the nucleus was arranged in rows.The cells in the fibrocartilage area were oval,and the closer to the bone,the larger the volume,there is a distinctly visible blue line between the calcified fibrocartilage areas and the uncalcified fibrocartilage area,the " tidemark ".The collagen fibers are aligned neatlyunder polarized light.The JUMP-1D,3D,5D groups,JUMP-p381 D,3D,and 5D days groups are basically the same as the CON group.Changes in cell density and thickness of fibrocartilage:The results of changes in cell density of patella tendon showed that there was no significant difference in JUMP-1D group compared with CON group,and the cell density of JUMP-3D group was significantly decreased(p<0.01),and the cell density of JUMP-5D group was decreased,there was a significant difference(p < 0.01).There was no significant difference in the JUMP-p381 D group compared with the JUMP-1D group.There was no significant difference in the JUMP-p383 D group compared with the JUMP-3D group.There was no significant difference in the JUMP-p385 D group compared with the JUMP-5D group.The results of changes in cell density of the patella-patella tendon junction showed that there was no significant difference in the JUMP-1D group compared with the CON group,and there was no significant difference in the JUMP-3D group,and there was no significant difference in the JUMP-5D group.There was no significant difference in the JUMP-p381 D group compared with the JUMP-1D group.There was no significant difference in the JUMP-p383 D group compared with the JUMP-3D group.There was no significant difference in the JUMP-p385 D group compared with the JUMP-5D group.The results of changes in the thickness of the fibrocartilage of patella-patella tendon junction showed that there was no significant difference in the JUMP-1D group compared with the CON group.There was no significant difference in the JUMP-3D group,and there was no significant difference in the JUMP-5D group.There was no significant difference in the JUMP-p381 D group compared with the JUMP-1D group.Immunohistochemistry results:The results of IL-1β in patella tendon showed that compared with CON group,IL-1βexpression in JUMP-1D group increased significantly,there was significant difference(p<0.05),and IL-1β expression in JUMP-3D group increased significantly.Significant differences(p < 0.05),there was no significant difference in the JUMP-5D group.There was no significant difference in the JUMP-p381 D group compared with the JUMP-1D group.There was no significant difference in the JUMP-p383 D group compared with the JUMP-3D group.There was no significant difference in the JUMP-p385 D group compared with the JUMP-5D group.The changes of IL-1β in the patella-patella tendon junction showed that compared with the CON group,the IL-1β expression in the JUMP-1D group group compared with the JUMP-5D group.There was no significant difference about the p38 MAPK in all groups about the patella tendon and patella-patella tendon junction.Conclusions:1 After the high-intensity exercise,there is no effect about the cell density and the thickness of the fibrocartilage of patella tendon and patella-patella tendon junction in the short term.2 The inflammatory response of tendon and tendon check point in the acute phase after intensive exercise was inconsistent,and p38 MAPK does not affect the inflammatory response in the acute phase.
