34 18-week-old female New Zealand white rabbits were randomly divided into control group(CON,n=4);jump 1 day(n=5),3 days(n=5),5 days group(n =5),referred to as J1,J3,J5;p38 inhibitor was injected after jumping 1 day(n=5),3 days(n=5),and 5 days(n=5),referred to as J1 P,J3P,J5 P.Inhibitor SB203580 was injected intraperitoneally after each training session.Each day,there are 10 groups for 150 qualified jumps,15 for each group.After the end of the other groups except the fourth group and the sixth group,the rabbit rested for 3 minutes in situ.After the end of the fourth group and the sixth group,the rabbit rested for 6 minutes in situ,and the feeding time was prohibited.After 24 hours of training,the samples were sacrificed(both ends of the sputum),and the paraffin sections were subjected to H&E and immunohistochemical treatment,and images were collected under the microscope for data analysis.RESULTS:(1)The PPTJ in healthy conditions was lower than the TPTJ in terms of cell density and collagen fiber arrangement,and the original storage of IL-1βand IL-6 in the two sites was larger.difference.(2)J1 group,the TGF-β1 of PPTJ was significantly lower than that of the CON group.At this time,the fibrosis reaction function and the performance of the two parts may have a gap,but the content of TGF-β1 in the two parts of the J1 P group was extremely significant.The sexual difference disappeared,the relative value of TGF-β1 in the two parts was similar to that in the CON group,the thickness of the cartilage band of TPTJ was abnormally increased,and the thickness of the cartilage band of PPTJ was abnormally reduced.At this time,the TPTJ with stronger adaptability was worse than histologically.PPTJ.(3)J5 group,TPTJ was still superior to PPTJ in histology,and the difference in the inflammatory response ability between PPTJ and TPTJ was narrowed.The gapbetween the mechanical properties of the two stops at one day gradually disappeared with the accumulation of load.After injection of p38 inhibitor,the IL-1β content of PPTJ decreased significantly,the collagen arrangement of TPTJ was disordered,the gap was enlarged,and histological performance was worse than PPTJ.Conclusions:(1)Under the same high-intensity short-term load,the adaptability of PPTJ to stress stimulation is weaker than that of TPTJ.(2)Under the high-intensity load of this experiment,the activation of p38 MAPK will affect the fibrosis process of PPTJ,thus affecting the mechanical properties of PPTJ.(3)Under the five-day load in this experiment,the activation of p38 MAPK affects the content of IL-1β in PPTJ,which affects the related inflammatory response of PPTJ.
