Objective: Research of primary Hypertension(Essential Hypertension,EH)patients peripheral blood lymphocyte subsets changes and folinic acid(with methylene reductase C677 T gene polymorphism,investigate folinic acid(methylene reductase(MTHFR)C677T gene polymorphism in patients with primary Hypertension immune inflammatory mechanisms,the influence of for primary Hypertension provide theoretical basis for individualized treatment and drug targeted therapy.Methods:Random collection in November 2016 to December 2018 in Yi jishan hospital geriatric medicine 41 patients with essential hypertension(eh)patients in the hospital as hypertension group,23 of men and 18 women,mean age 69.95 + /-6.94 years old.During the same period,40 inpatients without hypertension were treated as the control group,including 20 males and 20 females,with an average age of 67.40 + /-6.97 years.The hypertension group was divided into the wild-type(CC)group and the mutant(CT/TT)group according to the results of genetic testing,among which 20 patients were in the CC group,including 12 males and 8 females,with an average age of 71.55 + /-7.23 years;CT/TT group included 21 patients,including 11 males and 10 females,with an average age of 68.43+ /-6.45 years.To collect all the research object of peripheral blood in the morning,using flow cytometry to determine total peripheral blood T lymphocyte percentage(CD3 +),T lymphocytes(CD3 +,CD4 +),T inhibit lymphocyte percentage(CD3 + CD8 +),T cell/T suppressor cell percentage(CD4 + / CD8 +)ratio,NK cells(CD3 + CD16 + 56)percentage,B lymphocyte percentage(CD3-CD19 +);Fluorescence staining in situ hybridization was used to detect the C677 T locus of MTHFR gene in peripheral blood of patients with hypertension in the morning,and the genotypes were obtained,among which CC type was wild type,CT type was heterozygous mutant,and TT type was homozygous mutant.Results:1.The percentage of total T lymphocytes(CD3+),percentage of T helper lymphocytes(CD3+ CD4+)and percentage of T inhibitory lymphocytes(CD3+CD8+)in the peripheral blood of the hypertension group was lower than that of the control group(P<0.05).The percentages of NK cells(cd3-cd16 +56+)and B lymphocytes(cd3-cd19 +)were higher than those in the control group(P<0.05).2.The percentage of peripheral blood t-helper lymphocytes(CD3 +CD4+)and the ratio of t-helper/t-suppressor cells(CD4+/CD8+)in the CT/TT group were higher than those in the wild-type group(P<0.05).Conclusion : 1.Peripheral blood lymphocyte subsets of patients with essential hypertension were significantly different from those of patients without hypertension,suggesting the existence of immune dysfunction in patients with essential hypertension.2.The expression of peripheral blood lymphocyte subgroup in the MTHFR C677 T gene mutation group was significantly different from that in the wild group,suggesting that gene polymorphism may play a role in the immune inflammatory mechanism of essential hypertension. |
