Objective To study whether the C677T polymorphism of methylenete-trahydrofolate reductase (MTHFR) gene is associated with primary hyperuricemia and gout of Han nationality in eastern coast of Shandong Province.Methods MTHFR gene C677T polymorphism in 253 subjects of Han nationality in eastern coast of Shandong Province were examined by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) method,of which there are 153 subjects with high uric acid levels and 100 normal subjects(CON 100 cases,female 25,male 75,mean age43.42+12.34Y). The 153 subjects were divided into two groups :merely hyperuricemia group (UA-1 96 cases,female 24 ,male 72,mean age 43.06+12.11Y)and hyperuricemia associated with gout group(UA-2 57 cases,female 11,male46,mean age 45.28+9.3Y) by with or without gout attacks. Combined by higher concentration agarose gel electrophoresis,the allele and genotype among each group were compared. The relationship between the clinical biochemical results , gene polymorphism and the mobility of hyperuricemia were studied by Logistic regression analysis.Results (1)There are T ,C allele and CC,CT,TT genotype in the 253 cases.The mutational T allele frequency is 39% in group CON and 55% in group UA, which are significantly different(P<0.01) .In group CON, there is not difference of genetype frequency between male and female ,and so is group UA.(2)There are not significantly difference between sex,age,body mass index(BMI),blood pressure,fasting blood glucose,creatinine,urea nitrogen,plasma uric acid , triglyceride and cholesterol in group UA-1 and group UA-2 (P>0.05).(3) The frequencies of both allele T and genetype TC+TT are significantly higher in group UA-1 and group UA-2 than in group CON (P<0.05).But there is not significantly difference between group UA-1 and UA-2. (4)In group UA and group CON ,the uric acid levels of TT genetype are significantly higher than TC, CC genetypes(P<0.05).(5) Logistic regression analysis suggests that the allele T ,systolic blood pressure and BMI are risk factors of hyperuricemia.
