Background:The cornea is a dense tissue composed of an epithelial cell layer,the Bowman layer,the stromal layer,Descemet’s membrane,and an endo-thelial cell layer.The corneal epithelium is a protective barrier to the out-ermost layer of the cornea.It plays a vital role in resisting external dam-age factors such as bacterial damage and maintaining corneal transpar-ency and vision.Epithelium defects can be caused by adverse events of physical,chemical,and pathological factors,such as some systemic dis-ease,dry syndrome and diabetes.In general,a small area of corneal epithelial wound can be repaired within 24 hours without leaving scars.However,if further wound occurs,it may cause serious complications such as ulcers,perforations,endophthalmitis,and blindness.The healing of corneal epithelial wounds involves a number of concerted events in-cluding cell migration,proliferation,adhesion and differentiation with cell layer stratification.It involves growth factor、cytokine and extracel-lular matrix(ECM)signal-mediated interactions at the wound site to re-establish epithelial integrity and restore corneal homeostasis.Growth factors have been widely used in patients with corneal epithelial wound.Among them,basic fibroblast growth factor(b-FGF)has been proven to be an effective factor to promote the healing of corneal epithelium.In this article,we focus on the efficacy of the Specific Binding Recombinant Fi-broblast Growth Factors(v-bFGF)on corneal epithelial wound.This new growth factor has a unique collagen linker that has been shown to bind stably to collagen,while the Bowman layer and the corneal stroma are mainly composed of collagen fibers,providing a convenient condition for our research.Purpose:The aim of the study is to research treatment effect of v-bFGF on pro-moting corneal epithelial wound healing and compare the effect of v-bFGF and b-FGF.Methods:Sixteen Japanese rabbits were randomly divided into v-bFGF group,b-FGF group,PBS group and normal saline group.Each group has 4 eyes from 4 rabbits.The eyes were created a model of corneal epithelial wound.Eye drops 4 times a day,which started 3 hours after operation.The corneal epithelial healing rate of the experimental group and the con-trol group were observed.Compared with PBS group and NaCl group,a difference in the healing ability of corneal epithelial injury was observed and analyzed in v-bFGF group and b-FGF group.At the same time,a difference of the healing rate of corneal epithelium was observed and analyzed between the v-bFGF group and the b-FGF group.Result:By experimentally studying the effects of different drug treatments on the corneal epithelial injury model in rabbits,the novel specific binding re?combinant fibroblast growth factor(v-bFGF)was inducted and purified from E.coli BL-21 expressing v-bFGF,which was defined a good thera-peutic effect on corneal epithelial wound.The therapeutic effect of v-bFGF group and b-FGF group was significantly better than that of PBS control group and normal saline control group.Among them,v-bFGF was more effective than b-FGF to promote the healing of corneal epithe-lial wound.The v-bFGF group and the b-FGF group were significantly different from the saline group(p<0.05).The v-bFGF group and the b-FGF group were statistically significant(p<0.05).p<0.05),the v-bFGF group was statistically significant compared with the PBS group(p<0.05),while the b-FGF group was not statistically significant compared with the PBS group(p>0.05).There was no statistically significant difference be-tween the saline group and normal saline group(p>0.05).Conclusion:v-bFGF was more effective on promoting repair of corneal epithelial damage than b-FGF. |
