Background:Epithelial-mesenchymal transition(EMT)and chondrioditin ulfate pro-teoglycans 4(CSPG4)have been found in various fields of malignant tumors and systemic diseases.However,there are relatively few studies in oral squamous cell carcinoma(OSCC).This study is to detect the protein of EMT-related transcription factors SNAIL and TWIST 1 and CSPG4 in OSCC tissues and adjacent tissues of OSCC.The mRNA level,the relationship between EMT-related transcription factors SNAIL,TWIST 1 and CSPG4 and OSCC were presumed,which provided a theoretical basis for the later study of the specific mechanism of CSPG4 in OSCC.Objective: To detect the expression of CSPG4,SNAIL and TWIST 1 in OSCC tissues by immunohistochemistry and RTPCR,and to explore the relationship between CSPG4,SNAIL,TWIST 1 and OSCC,and provide theoretical basis for early diagnosis and prevention.METHODS:(1)RNA of CSPG4,SNAIL and TWIST 1 in cancer tissues and paracancerous tissues of OSCC patients were extracted,and the expression levels of mRNA in the above items were verified by real-time fluorescent quantitative PCR,and CSPG4,SNAIL and TWIST 1 were further analyzed in OSCC.The mechanism of action.(2)Immunohistochemical technique was used to detect the expression of CSPG4,SNAIL and TWIST 1 in cancer tissues and paracancerous tissues of patients with OSCC.Results:(1)CSPG4 was highly expressed in OSCC cancer tissues,and the staining score was higher than that in adjacent tissues(P<0.001).In cancer tissues,CSPG4 protein was mainly expressed in the cytoplasm and membrane of cancer cells.The membrane was deeply stained,the keratinized beads were not stained,and the normal tissue adjacent to the cancer,CSPG4 protein,was expressed in a small amount in the normal adjacent epithelium.The protein expression of CSPG4 was not correlated with age,ethnicity,T stage and OSCC differentiation(P>0.05),and was associated with gender,lymph node metastasis and clinical stage.The expression of SCPG4 protein in male OSCC patients was higher than that in females(P< 0.05),OSCC with lymph node metastasis group was higher than non-metastasis group(P<0.01),clinical stage III-IV was higher than stage I-II(P<0.05),in male patient group,metastasis group and stage III-IV group The positive rate was higher and the staining intensity was stronger.The positive rate of female patient group,non-metastasis group and stage I-II group was lower,and the staining intensity was weaker.The expression of CSPG4 mRNA was not correlated with age,ethnicity,T stage and OSCC differentiation(P>0.05),and was associated with gender,lymph node metastasis and clinical stage.The expression of male patients was higher than that of females(P<0.05).The metastatic group height was higher(P<0.05),and the clinical stage III-IV phase was higher than the I-II phase(P<0.05).(2),Snail protein is mainly expressed in cancer nest tissue in OSCC tissues,cytoplasm and membrane of cancer cells,deep staining of membrane,keratinized beads are not stained,in normal tissues adjacent to cancer,Snail protein is mainly expressed in normal epithelium The base layer is shallower toward the keratinized layer in the shallow direction.The Snail protein staining score of OSCC tissues was higher than that of adjacent normal tissues(P<0.01).Combined with clinical case information,the expression of Snail protein was not correlated with gender,age,ethnicity,T stage and OSCC differentiation(P>0.05),and was associated with lymph node metastasis and smoking and drinking(P<0.05).Real-time PCR was used.The expression of Snail mRNA at the transcriptome level was detected by qPCR.The relative expression of Snail mRNA in OSCC tissues was higher than that in adjacent tissues.The expression of SNAIL mRNA was not correlated with age,ethnicity,T stage and OSCC differentiation(P>0.05),and was associated with lymph node metastasis,smoking and drinking.The metastatic group was higher than the non-metastasis group(P<0.05).The lymph node metastasis of oral squamous cell carcinoma was higher than that of no lymph node metastasis group.The expression level of smoking group was higher than that of non-smoking group(0.693-4.901).The expression level of drinking group was higher than that of non-drinking group,and there was no obvious among other groups.Statistical differences.(3)TIWST1 protein is mainly expressed in cancer nest tissue in OSCC tissues.The membrane is deeply stained,and the central keratinized beads are not stained.In the normal tissues adjacent to the cancer,it is mainly expressed in the normal epithelial basal layer,gradually toward the keratinized layer.Lightening,overall staining is lighter,and the staining score of OSCC tissue is higher than that of adjacent normal tissues.Combined with clinical case information,TIWST1 protein expression was not associated with patient ethnicity,alcohol consumption,gender,T stage,age and OSCC differentiation(P>0.05),and was associated with lymph node metastasis and smoking(P<0.05).Real-time quantitative PCR(qPCR)was used to detect the expression of TWIST 1 mRNA at the transcriptome level.Combined with clinical case information,the expression of TWIST 1 mRNA was not associated with patient ethnicity,alcohol consumption,gender,T stage,age and OSCC differentiation(P>0.05),and lymph node.Metastasis and smoking were correlated(P<0.05),and the expression level in OSCC cancer tissues was higher than that in adjacent tissues.Conclusion:(1)The large expression of CSPG4 in OSCC indicates that there is a positive regulation between CSPG4 and OSCC,which provides a possibility for early diagnosis and prognosis of OSCC,and even provides a targeted therapeutic strategy in the treatment of OSCC.(2)The EMT-related factors SNAIL and TWIST 1 are highly expressed in OSCC,suggesting that the relationship between EMT and OSCC is very close.(3)SNAIL and TWIST 1 were associated with OSCC lymph node metastasis,suggesting that SNAIL and TWIST 1 and even EMT positively regulate the lymph node metastasis of OSCC.(4)Both SNAIL and TWIST 1 are associated with smoking habits in OSCC patients,suggesting that smoking may be one of the causes of EMT.(5)As one of the upstream regulatory proteins of EMT,CSPG4 may be an important factor in inhibiting the development of EMT and OSCC.
