BackgroundTransitional Cell Carcinoma of Bladder(TCC)is one of the most common urinary tract tumors,the morbidity and mortality of which account for the first place in urinary tract tumors,and it has a gradual upward trend,bringing a lot of trouble and pain to patients and their families.In recent years,a large number of literatures have shown that Wnt signaling pathway plays an important role in the occurrence,development and high recurrence of bladder cancer in bladder transitional cell carcinoma.ObjectiveThis study will investigate the relationship between Wnt1,Dickkopf-1,β-catenin and the depth(stage),degree of differentiation(pathological grade),diameter,recurrence,metastasis and prognosis of transitional cell carcinoma of the bladder.To explore its role in the occurrence,invasion,metastasis and recurrence of bladder transitional cell carcinoma,to find tumor markers for judging the malignant degree and prognosis of bladder transitional cell carcinoma,and to provide theoretical basis and target for early screening and post-treatment.Methods1、Collecting 60 cases of pathological tissue confirmed by clinical diagnosis and pathology in our hospital(including 20 cases in T2a stage,20 cases in T2b stage,20cases in T3 stage),20 cases of normal bladder tissue,collect serum samples from patients with bladder cancer and normal subjects simultaneously.Western blot and RT-qPCR were used to detect the expression of Wnt1/Dickkopf-1/β-catenin protein in normal bladder tissue,bladder cell and serum.2、Analyze the relationship between the expression of Wnt1/Dickkopf-1/β-catenin and bladder cancer,depth of myometrial invasion,pathological grade,age,lymph node metastasis,tumor diameter,and recurrence.Analyze the correlation of Wnt1/Dickkopf-1/β-catenin expressionResults1、Wnt1 andβ-catenin mRNA and protein are expressed in normal bladder mucosa group,T2a group,T2b group and T3 group.The expression levels of Wnt1 andβ-catenin mRNA and protein in bladder tumor tissues are significantly higher than those in normal control group.The deeper the infiltration is,the higher the expression level will be,and the difference among the four groups is statistically significant(P<0.05).2、Dickkopf-1 mRNA and protein were expressed in normal bladder mucosa group,T2a group,T2b group and T3 group.The expression level of Dickkopf-1 mRNA and protein in bladder tumor tissues was significantly lower than that in normal control group,The deeper the infiltration is,the lower the expression level will be,and the difference among the four groups is statistically significant(P<0.05).3、There is no significant difference in the expression of Wnt1 andβ-catenin mRNA and protein between male and female in bladder cancer tissues(P>0.05).The expression level of Wnt1 andβ-catenin in the≥60 years old group is higher than that in the<60 years old group,and the difference is significant(P<0.05);the expression of lymph node metastasis group is higher than that of no lymph node metastasis group,the difference is significant(P<0.05);the tumor diameter≥3cm group was higher than the tumor diameter<3cm group,the difference is Significant(P<0.05);the expression of recurrence group was higher than that of non-recurrence group at 1year follow-up(P<0.05);the expression level in G3 group was higher than that of G1G2 group,the difference is significant.(P<0.05).4、There is no significant difference in the expression of Dickkopf-1 mRNA and protein between male and female in bladder cancer tissues(P>0.05).The expression level of Dickkopf-1 in the≥60 years old group was lower than that in the<60 years old group,and the difference was significant(P<0.05);the expression of lymph node metastasis group was lower than that of no lymph node metastasis group(P<0.05);the expression of tumor diameter≥3cm group was lower than tumor diameter<3cm group,the difference was significant(P<0.05);The expression of recurrence in the recurrence group was lower than that in the non-recurrent group(P<0.05).The expression in the G3 group was lower than that in the G1G2 group(P<0.05).5、The serum levels of Wnt1 andβ-catenin in patients with bladder cancer were higher than those in the normal control group.The differences were statistically significant(P<0.05);the Dickkopf-1 protein level was lower than the normal control group,and the difference was statistically significant(P<0.05).6、There is a negative correlation between the expression of Wnt1 and Dickkopf-1 mRNA in bladder cancer,and the correlation coefficient was:r=-0.760,P=0.000.There was a negative correlation between the expression ofβ-catenin and Dickkopf-1 mRNA,and the correlation coefficient was:r=-0.763,P=0.000.There was a negative correlation between the expression of Wnt1 and Dickkopf-1 protein,and the correlation coefficient was:r=-0.828,P=0.000.There was a negative correlation between the expression ofβ-catenin and Dickkopf-1 protein,and the correlation coefficient was:r=-0.696,P=0.000.Conclusions1、The expression of Wnt1 andβ-catenin in bladder tissue of bladder tumor patients increases with the depth of tumor invasion.The expression of Dickkopf-1decreased with the increase of tumor infiltration depth,suggesting that Wnt1,β-catenin.Dickkopf-1 may be involved in the pathogenesis of bladder tumors,and its expression level may reflect the malignant degree of bladder cancer to some extent.2、The expression levels of Wnt1,β-catenin and Dickkopf-1 are correlated with age,tumor diameter,pathological grade,lymph node metastasis and recurrence,suggesting that the expression of the three may affect the prognosis of patients.3、There is a negative correlation among the expression of Wnt1,β-catenin and Dickkopf-1 in bladder cancer,suggesting that the expression loss of Dickkopf-1 may promote the abnormal expression of Wnt1 andβ-catenin.4、The expression levels of Wnt1,β-catenin and Dickkopf-1 in serum of patients with bladder cancer may be used as a new tumor marker to provide a basis for early diagnosis and treatment of bladder cancer.
