Objective: To investigate the expression differences of RHBDD2 and SPAG4 in colorectal cancer and adjacent normal tissues,and to explore the relationship between RHBDD2,SPAG4 and the clinical data characteristics,pathological features and prognosis of colorectal cancer.Method: A total of 87 specimens of patients with colorectal cancer who underwent surgery from January 2010 to January 2018 in the Affiliated Hospital of Capital Medical University were selected to make tissue microarrays.The immunohistochemical method was used for comparative analysis.The expression differences of RHBDD2 and SPAG4 in tumor tissues and adjacent normal tissues were observed.To analyze the relationship between gene expression differences in tumor tissues and clinical data characteristics,pathological features and prognosis of patients,and to explore the clinical application value of RHBDD2 and SPAG4 in the diagnosis,treatment and prognosis of colorectal cancer.Results: 1.High expression rates of RHBDD2 and SPAG4 were 53.45% and 72.46% respectively in the samples of colorectal cancer tissues,and the differences were statistically significant compared with the paracancer normal tissues(chi-square =4.391,5.301,P < 0.05).2.The results of immunohistochemistry assay on tissue microarray showed that tumor diameter,Dukes’ stage,lymph node metastasis were significantly correlated with the high expression of RHBDD2(P < 0.05).And there was no significant difference with age,gender,pathological type,differentiation degree,the depth of infiltration and nerve invasion(P>0.05).The high expression of SPAG4 was significantly different from tumor diameter,Dukes’ stage and the depth of infiltration(P < 0.05),but not from age,gender,pathological type,differentiation degree and others.3.Kaplan-meier survival analysis was performed to analyze the relationship between RHBDD2,SPAG4 expression and the prognosis of CRC patients.The overall survival of the high-expression group was shorter than that of the low-expression group,and the difference was statistically significant(P < 0.05).4.Overall survival of colorectal cancer is correlated with tumor diameter,Dukes’ stage,lymph node metastasis,the depth of infiltration and vessel cancer embolus(P < 0.05).Conclusions: 1.RHBDD2 and SPAG4 expression may be related to the occurrence and development of colorectal cancer.2.The high expression of RHBDD2 may be correlated with the diameter,as well as the lymph node metastasis and stage of the tumor.SPAG4 high exprssion may be correlated with tumor diameter,the depth of infiltration and stage.3.High expression of RHBDD2 and SPAG4 suggested poor prognosis and shortened overall survival of colorectal cancer.4.Overall survival of colorectal cancer is correlated with tumor diameter,stage,lymph node metastasis,depth of infiltration and vessel cancer embolus.
