Objective:Crataegi Fructus Carbonnisatus is a processed product of Crataegi Fructus Carbonnisatus.After processing,it has the effect of astringency,hemostasis,astringency and diarrhea.At present,there is no clear specification for the processing of Crataegi Fructus Carbonnisatus and lack of accurate quantitative indicators,resulting in uneven quality of Decoction pieces,which has a great impact on the production and application of Crataegi Fructus Carbonnisatus.In order to guide the production of Crataegi Fructus Carbonnisatus more effectively,the L9(34)orthogonal design method was adopted in this study.The changes of appearance and content of active ingredients were taken as the evaluation indexes.The optimum processing technology of Crataegi Fructus Carbonnisatus was optimized by selecting three factors:frying temperature,frying time and turning times,so as to ensure the consistency of its appearance and internal quality,and the processing method was stable and feasible.Secondly,pilot-scale production was carried out,and quality standards were formulated accordingly to provide quality control basis for the production and clinical application of Crataegi Fructus Carbonnisatus,and to promote the standardization and standardization of the processing and production of traditional Chinese medicine charcoal.Methods:1.Literature study:Summarize and summarize the research contents of Crataegi Fructus Carbonnisatus in recent years,including its historical evolution,processing technology,chemical composition changes,pharmacological effects,clinical application and so on.2.Technological research:According to the Pharmacopoeia method,Crataegi Fructus Carbonnisatus slices from different producing areas were checked for quality,and the qualified and practical Crataegi Fructus Carbonnisatus slices were screened out;Processing technology research:The orthogonal experimental design method was used to study the technological parameters,in order to directly influence the processing time,temperature and turnover times of processed products,and three water samples were designed through pre-experiment.Ping.The appearance,color difference and content of active ingredients of Crataegi Fructus Carbonnisatus decoction pieces are correlated with their processing degree,which can represent the processing quality of Crataegi Fructus Carbonnisatus Decoction pieces.Based on this,the best processing technology of Crataegi Fructus Carbonnisatus can be judged.3.Quality standard:To determine the indexes of the samples of Crataegi Fructus Carbonnisatus produced according to the best processing technology,including the methodological investigation of TLC,the determination of moisture,ash and extracts,the limit inspection of heavy metals and agricultural residues,the formulation of the standards of property,taste and meridian tropism,function and main treatment,usage and dosage,and storage.4:Pharmacodynamic study:The effects of Crataegi Fructus Carbonnisatus on coagulation time and bleeding time in mice were investigated by capillary method and Tail-cutting method.The plasma prothrombin time,activated partial thromboplasti time,thrombin time and fibrinogen were measured by coagulation analyzer to explore the mechanism of coagulation and hemostasis.Results:Through comprehensive evaluation,the optimum processing parameters of Crataegi Fructus Carbonnisatus Carbon Decoction were as follows:processing temperature was 220 C,processing time was 15 min,and turnover frequency was 8 r/min.The quality standards and drafting instructions of Crataegi Fructus Carbonnisatus were formulated.Through pharmacodynamics study,Crataegi Fructus Carbonnisatus decoction can significantly shorten the time of blood coagulation and bleeding in mice,indicating that Crataegi Fructus Carbonnisatus has hemostatic and coagulative effects.The Crataegi Fructus Carbonnisatus decoction can significantly shorten PT,APTT,TT and increase the number of FIB in mice by coagulation analyzer.Conclusion:The optimum processing technology of Crataegi Fructus Carbonnisatus has been worked out through this research,and the processed products are uniform and stable from appearance to internal quality.The quality standard of Crataegi Fructus Carbonnisatus drafted has been checked and accepted,which can be used as the basis for quality control of Crataegi Fructus Carbonnisatus.Through pharmacodynamic experiments,the pharmacological effect of Crataegi Fructus Carbonnisatus was proved,and the traditional use of Crataegi Fructus Carbonnisatus for hemostasis was verified.
