Purpose:1.Studying the law of electroacupuncture for insomnia;2.Polysomnography(PSG)in the form of EEG and EMG monitoring was used as a gold indicator to analyze the difference in therapeutic effects between single acupoints and acupoint compatibility;3.To clarify the regulation of endogenous sleep-related molecular substances in peripheral blood of insomnia rats by electroacupuncture stimulation.To explore the therapeutic effect and mechanism of electroacupuncture at single point and acupunct compatibility with acupoints on insomnia.The principle of selecting acupuncture points for clinical acupuncture treatment of insomnia provides a certain experimental basis for the regulation of endogenous molecules in the body.Method:A total of 120 male SD rats were randomly assigned.There were 60 rats in the electrophysiological group,10 in each group,and 60 non-electrophysiological groups,10 in each group.The electrophysiological group and the non-electrophysiological group were the same in 6 groups,namely normal group,model group,therapy group,therapy group incloud HT7 group,GV20 group,SP6 group and acupoint compatibility group "HT7-GV20-S06".1.The electrophysiology group had to carry out electrode embedding surgery before the modeling.After the rats were placed in a self-made plexiglass cage,they were fed for one week and then molded.2.The model of insomnia rat was duplicated by intraperitoneal injection of p-chlorophenylalanine(PCPA)once/1d for 2 days.After rats in the first intraperitoneal injection of 28~30h,the circadian rhythm of rats disappeared,and the daytime activities were significantly different from the blank control group,that is,the model was successfully reproduced.3.Treatment was started on the third day of modeling.The electrophysiological group began to monitor electrophysiology on the first day of treatment,and was treated for 7 days.The non-electrophysiological group was treated for 4 days.After the end of the treatment,materials were taken and ELISA was used.ELISA was used to detect the levels of melatonin,prostaglandin D2,brain-derived neurotrophic factor,interleukin-1β,and corticosterone in peripheral blood of rats in each group.Result:1.PSG analysis showed that the awakening time of the model group rats on the 4th day was significantly longer than that of the normal group rats(p<0.05).Compared with the model group,the awakening time of the treatment group rats were both Significantly decreased(p<0.05),and the awakening time of the combination group was significantly less than the other three treatment groups(p<0.05).2.The content of MT,PGD2,BDNF,IL-1β and CORT in the peripheral blood of each group of rats:The ratio of MT,PGD2,BDNF and IL-1β in the model group was significantly lower than that in the normal group and the model group(p<0.05),CORT content was significantly increased(p<0.05);compared with the model group,the serum levels of MT,PGD2,BDNF,and IL-1β were significantly higher in the treatment group,and the combined points were superior to other treatment acupoint groups.(p<0.05),the CORT content in the treatment group decreased significantly and was superior to other treatment acupoint groups(p<0.05).Conclusion:1.Electroanalysis for insomnia has significant therapeutic effects both in terms of electrophysiological level and molecular level.2.Electroacupuncture at one point and compatibility with acupoints relieved the insomnia in rats,and the combined acupoint group was superior to single acupoint group in the treatment of insomnia.
