Objective: To research the effect of BDNF and NGF in theintracerebral hemorrhage rats’brain with transplantation of BMSCscombining electroacupuncture of Baihui (GV20) and Dazhui (GV14).Methods: Establishing intracerebral hemorrhage animal models byinjecting collagenase and heparin sodium into Adult SD rats’Caudate-Putamen by stereotaxis instrument, and then the rats wererandomly divided into normal saline group(Contra group),Bone marrowmesenchymal stem cells transplantation group (BMSCs group) andElectroacupuncture combining Bone marrow mesenchymal stem cellstransplantation group (Ea BMSCs group). Then Rats of each group weredivided into5subsets according to re-feeding time (1dã€3dã€5dã€7dã€14d.6rats in each subet). Graft were transplated into the intracerebral hemorrhageanimal models2hours later after the establishment of animal models bystereotaxis instrument. Rats of Ea BMSCs group were stimulated withelectroacupuncture on Baihui (GV20) and Dazhui (GV14) after2h cellstransplanted and one time a day. At different time points of re-feeding(1dã€3dã€5dã€7dã€14d) execute and remove the brain, then the expressionlevel of BDNF mRNA and NGF mRNA was observed by reverse transcriptase polymerase chain reaction(RT-PCR). After2h of animalmodel established, we make a Neurological Severity Score (NSS) andrecord it, then record it one time each time points until death of SD rats.Result:1. The Ea BMSCs group’s Expression of BDNF mRNA was higherthan the expression of Contra group at the same time point (1d,7d,14d),the difference was statistically significant, P<0.05. The Expression ofBDNF mRNA of BMSCs group was higher than the expression of Contragroup at the same time point of7d, the difference was statisticallysignificant, P<0.05. The Expression of7d was higher than it of3d and5din the Ea BMSCs group, the difference was statistically significant,P<0.05.2. The Ea BMSCs group’s Expression of NGF mRNA was higherthan the expression of Contra group at the same time point (3d,5d,7d,14d),the difference was statistically significant, P<0.05. The BMSCs group’sExpression of NGF mRNA was higher than the expression of Contra groupat the same time point of7d, the difference was statistically significant,P<0.05.3. The NSS score difference at the same time point beforetransplantation was not satistically significant between every experimentalgroup (Contra group, BMSCs group and Ea BMSCs group). The NSS scoreat the time lower than it of next time point in the same experimental grouptransplantation, it was statistically significant, P<0.05. The BMSCsgroup’s NSS score lower than the score of Contra group at the same timepoint of5d,the difference was statistically significant, P<0.05. The Ea BMSCs group’s NSS score lower than it of Contra group at the same timepoint (3d,5d,7d,14d), the difference was statistically significant,P<0.05.conclusion: BMSCs transplantation combining electroacupuncturestimulation can enhance expression of BDNF and NGF after intracerebralhemorrhage, more improve neurologic deficits symptoms of the cerebralhemorrhage SD rats than simple BMSCs transplantation. |
