Study On The Expression Of MiRNAs In Patients With Intracranial And Extracranial Artery Stenosis

Objective:The risk factors of ischemic stroke patients and the expression levels of miR-17-5p、miR-27b-3p、miR-92a-3p、miR-125a-5p、miR-186-5p and miR-208b-3p in serum were detected,to explore the correlation between them and the stenosis of intracranial and extracranial arteries,and the feasibility of serum miRNAs as potential biomarkers for intracranial and extracranial artery stenosis.Methods: 99 patients with ischemic stroke were chosen.According to the results of DSA,they can be divided into four groups(intracranial artery stenosis group,extracranial artery stenosis group,intracranial and external artery stenosis group,control group).Compare the expression between the artery stenosis group and control group in related risk factors as age、male、smoking、hypertension、diabetes、neutrophils/lymphocyte ratio(NLR)、uric acid(UA)、cystatin C(Cys-c)、total cholesterol(TC)、triglycerides(TG)、high-density lipoprotein cholesterol(HDL-C)、low density lipoprotein(LDL-C),etc.The correlation between them and the degree of intracranial and external arterial stenosis was also analyzed.Total RNA was extracted from serum,Real-time quantitative PCR(RT—PCR)was used to detect expression of miR-17-5p、miR-27b-3p、miR-92a-3p、miR-125a-5p、miR-186-5p and miR-208b-3p.SPSS 22 statistical software was used for statistical analysis of the relative expression levels of miRNAs.Numeration data was expressed as mean ± standard deviation(X±S).T-test or nonparametricrank sum test was used for comparison between the two groups,and one-way anova or nonparametric rank sum test was used for comparison between multiple groups.P<0.05 was considered as statistically significant.Then,patients with artery stenosis were divided into slight stenosis group,moderate stenosis group and severe stenosis group according to the degree of stenosis.The correlation between the expression level of miRNAs and the degree of stenosis was analyzed.Results:(1)Artery stenosis groups of age 、 male,smoking、hypertension、UA、Cys-c、TC、LDL-C、Apo A1、Apo B,respectively,compared with control group was significant(P < 0.05).Age、TC and Apo A1 were independent risk factors for intracranial and external arterial stenosis(P <0.05).(2)Compared with the control group,there were significant differences in UA 、 LDL-C 、 Apo A1 and Apo B in the groups with different degrees of intracranial and external artery stenosis(P < 0.05).(3)The expression levels of serum miR-17-5p、miR-27b-3p、miR-92a-3p and miR-186-5p in the group of artery stenosis were significantly higher than those in the control group,while the expression levels of miR-125a-5p in the group of artery stenosis were significantly lower than those in the control group,with statistically significant differences(P <0.05).(4)miR-17-5p in the severe stenosis group was significantly higher than that in the mild stenosis group(P < 0.05).The expression level of miR-27b-3p in the severe stenosis group was higher than that in the mild stenosis group and the moderate stenosis group(P < 0.05).miR-92a-3p was statistically significant between the light,moderate and severestenosis groups(P < 0.05).miR-125a-5p in the severe stenosis group was significantly lower than that in the mild stenosis group(P < 0.05).miR-186-5p showed no significant difference between groups with different degrees of stenosis(P > 0.05).(5)The expressions of miR-17-5p、miR-27b-3p、miR-92a-3p and miR-186-5p in the group with intracranial and extracranial artery stenosis were higher than those in the group with intracranial artery stenosis and the group with extracranial artery stenosis,while the expressions of miR-125a-5p in the group with intracranial and extracranial artery stenosis were lower than those in the group with intracranial artery stenosis and the group with extracranial artery stenosis,but there was no statistically significant difference(P > 0.05).Conclusion:(1)Age、male、smoking、 hypertension、 UA、Cys-c、 TC、 LDL-C、 Apo A1、Apo B are risk factors of intracranial and extracranial artery stenosis.Age、TC and Apo A1 were independent risk factors for intracranial and extracranial arterial stenosis.(2)UA、TC、LDL-C、Apo A1、Apo B and the degree of intracranial artery stenosis were correlated.(3)The levels of serum miR-17-5p 、 miR-27b-3p 、 miR-92a-3p 、 miR-125a-5p and miR-186-5p are closely related to the intracranial and extracranial artery stenosis in patients with ischemic stroke.They are risk factors for intracranial and extracranial artery stenosis and may be potential biological markers for the diagnosis of intracranial and extracranial artery stenosis.(4)The relative expression levels of serum miR-17-5p 、 miR-27b-3p 、 miR-92a-3p and miR-125a-5p were correlated with the degree of intracranial and extracranialartery stenosis.(5)The expressions of serum miR-17-5p 、 miR-27b-3p 、miR-92a-3p、miR-125a-5p and miR-186-5p could not effectively distinguish the locations of intracranial and extracranial artery stenosis.
