| Objective:To investigate the reversal effect and mechanism of IGBR on TGF-β1-induced EMT model of human hepatoma cell lines HepG2,SMMC-7721 and SK-Hep 1.Methods:Human hepatoma cell lines HepG2,SMMC-7721 and SK-Hepl were induced EMT model by TGF-β1.Three cell lines morphological changes were observed under microscope,migration ability was detected by Wound Healing,invasion ability was detected by Transwell,the expression of E-cadherin,N-cadherin and Vimentin in the control group,model group and IGBR group was detected by Western blot and RT-PCR.Results:1.HepG2 cells;Compared with the control group,the cells in the model group were spindle shape and the number of cells increased.Wound Healing results showed that compared with the control group,the number of migratory cells in the model group increased;Compared with the model group,the number of migratory cells in the IGBR group decreased Transwell results showed that compared with the control group,the number of invasive cells in the model group increased;Compared with the model group,the number of invasive cells in the IGBR group decreased Western blot and RT-PCR results showed that compared with the control group,in the model group the expression of E-cadherin was decreased,and the expression of N-cadherin and Vimentin were increased;Compared with the model group,in the IGBR group the expression of E-cadherin was up-regulated,and the expression of N-cadherin and Vimentin were down-regulated.2.SMMC-7721 cells:Compared with the control group,the cells in the model group were long spindle shape and the number of cells increased.Wound Healing results showed that compared with the control group,the number of migratory cells in the model group increased;Compared with the model group,the number of migratory cells in the IGBR group decreased Transwell results showed that compared with the control group,the number of invasive cells in the model group increased;Compared with the model group,the number of invasive cells in the IGBR group decreased Western blot and RT-PCR results showed that compared with the control group,in the model group the expression of E-cadherin was decreased,and the expression of N-cadherin and Vimentin were increased;Compared with the model group,in the IGBR group the expression of E-cadherin was up-regulated,and the expression of N-cadherin and Vimentin were down-regulated3.SK-Hepl cells:Compared with the control group,the cells in the model group were long spindle shape and the number of cells increased.Wound Healing results showed that compared with the control group,the number of migratory cells in the model group increased;Compared with the model group,the number of migratory cells in the IGBR group decreased Transwell results showed that compared with the control group,the number of invasive cells in the model group increased;Compared with the model group,the number of invasive cells in the IGBR group decreased Western blot and RT-PCR results showed that compared with the control group,in the model group the expression of E-cadherin was decreased,and the expression of N-cadherin and Vimentin were increased;Compared with the model group,in the IGBR group the expression of E-cadherin was up-regulated,and the expression ofN-cadherin and Vimentin were down-regulated.Conclusions:IGBR inhibits the migration and invasion of human hepatoma cell lines HepG2,SMMC-7721 and SK-Hep1. |
