Objective:To study the effect of iridoid glucosides from Boschniakia rossica (IGBR) on serum marker enzymes and antioxidative enzymes of rats with hepatic preneoplasia induced by diethylnitrosamine (DEN) and investigatc the possible mechanism of its anti-carcinogenesis.Methods:Sixty male Wistar rats were randomly assigned into five groups, control group [saline-basal diet (BD)-partial hepatectecomy (PH)], model group [(DEN-acetylaminoflofluorene (AAF)-PH)], IGBR high dose group (DEN-AAF+500mg/kg IGBR-PH), IGBR medium dose group (DEN-AAF+250mg/kg IGBR-PH) and IGBR low dose group (DEN-AAF+125mg/kg IGBR-PH). Hepatic precancerous lesions in rats were induced by the modified Solt and Farher protocol. Rats from model and IGBR groups were treated with DEN and AAF to induce preneoplastic lesions of liver. Rats were administered with IGBR for six weeks, the activities of y-glutamy transpeptidase (y-GT), glutathione-S-transferase (GST), alanine trasaminase (ALT), aspartate amino-transferdse (AST), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-PX), as well as the contents of malondialdehyde (MDA) were detected by colorimetric method. The histological changes in DEN induced hepatic preneoplastic lesions of rats were observed by hematoxylin eosin (HE) staining.Result:IGBR obviously improved the mental state, color pattern, appetite, and weigh gain. In model group, the normal arrangement of the liver were lost, the structure of hepatic lobules disappeared and replaced by hepatocyte dysplasia foci and pseudolobules. IGBR proved the arrangement of liver with minor necrotic lesions and hepatocyte dysplasia foci. The serum y-GT, GST, ALT, AST activities and MDA level and were increased, and the serum SOD, CAT, GSH-PX activities were dereased in model group. The administration with IGBR reduced the activities of serumγ-GT, GST, ALT and AST, increased the serum SOD, CAT, GSH-PX activities, and decreased the serum MDA level in rats with hepatic preneoplastic lesions.Conclusion:IGBR exhibited an inhibitory effect on formation of hepatic preneoplastic lesions of rats, probably via its increased enzyme activities antioxdative activity.
