Object:Recently the role of long non-coding RNAs(lncRNAs)in carcinogenesis has been highlighted.However,there still remains largely unknown in lncRNAs involved in ovarian cancer(OC).In this study,we focused on lncRNA H19,and investigated the expression and functional role of H19 in OC.Methods:At the tissue level,30 specimens of ovarian cancer tissues and adjacent normal tissues were collected,the expression of H19 in specimens of ovarian cancer tissue sand adjacent normal tissues was measured by real-time fluorescence quantitative PCR(qPCR).And the relationship between the expression of H19 and the progress of disease,and the relationship between the expression of H19 prognosis indicators of patients were also analyzed.At the cellular level,expression of H19 in ovarian cancer SKOV3 cells was silenced by RNA interference(RNAi)and viral infection techniques.The cells were divided into control group(untreated SKOV3 cells),scramble group(SKOV3 cells infected with lentivirus carrying siRNA-Scramble),and experimental group(SKOV3 cells infected with lentivirus carrying siRNA-H19).Cell proliferation was assessed by cell counting Kit-8 and cell metastasis and cell apoptosis rate was observed by flow cytometry analyze.Results:Our results demonstrated that in 30 cases of tissue samples,in contrast to adjacent normal tissues,the expression of H19 in ovarian cancer tissue changed significantly,and each case had up-regulated in different degrees.And it was analyzed that high expression of H19 was associated with these prognostic factors such as patients’ FIGO stage(P<0.05)and surgical satisfaction(P<0.05),,but there was no significant correlation with age,histological grade,pathological type and preoperative serum CA125 expression level.With further investigation,we find that H19-silenced SKOV3 ovarian cancer cells,compared with the normal SKOV3 ovarian cancer cells,the expression of H19 decreased significantly.At the same time,the proliferation rate ofovarian cancer cells was decrease,and the apoptosis rate was increase.Conclusion:Our experiments confirmed that lncRNA H19 is overexpressed in both ovarian cancer tissues and cells,suggesting its potential carcinogenicity,and this overexpressed is associated with the progress of ovarian cancer,pointing out that its overexpressed often result in fast prognosis.In addition,the experiment also confirmed that silence H19 in vitro can inhibiting the proliferation of ovarian cancer SKOV3 cells,suggesting that H19 plays an important role in the occurrence and development of ovarian cancer.It is revealed that it may contribute to the malignant cell behavior of ovarian cancer.Our experiments provide new evidence for the development of ovarian cancer and provide new method for its treatment.With modern biotechnology we can use new therapy targeting at H19 on gene level.Moreover,H19 has a certain value in the evaluation of ovarian cancer prognosis.
