significance,to analyze the relationships between the expression of GATA3 and the expression of ER,PR,Her-2 and AR in breast cancer tissues and Tumor cell proliferation, and to further explore the significance of GATA3expression in different molecular types of breast cancer tissue.MethodA total of 209 samples with breast cancer undergoing surgical treatment at the Second Affiliated Hospital of Fujian Medical University from December 2010 to December 2016 were collected.According to the 2013 St.Gallen International Breast Cancer Conference on the development of molecular classification of breast cancer standards. The experiment divided 209 cases of breast cancer into Luminal type,Her-2 overexpression of breast cancer, triple negative breast cancer. The expression of GATA3,ER,PR,Her-2,AR and Ki-67 in breast cancer samples from209 cases of breast cancer were detected by immunohistochemistry(SP)And the proliferation index of Ki-67 in breast cancer tissues was calculated, with which to analyze the expression of GATA3 in breast cancer tissues and the relationship between the clinicopathological factors and ER,PR,Her-2,AR expression plus Ki-67 proliferation index.Based on that, the study attempts to further explore the significance of GATA3 expression in different molecular types of breast cancer.Result1 Correlation between GATA3 expression and clinicopath-ological factors in breast cancerThe positive rate of GATA3 in breast cancer was 60.8%(127/209 cases). The positive rate of GATA3 in moderately and strongly differentiated breast cancer was69.0%(78/113 cases),which was significantly higher than that in poorly differentiated breast cancer(51.0%,49/96),with significant difference(P<0.05).The positive expression rate of GATA3 in breast cancer with lymph node metastasis was 49.5%(46/93 cases),which was significantly lower than that in breast cancer without lymph node metastasis(69.8%81/116 cases),with significant difference(P<0.05).The positive rates of GATA3 in stage I,II and III breast cancer tissues were 71.6%(53/74),63.2%(55/87)and 39.6%(19/48)in TNM stage. With an increase in staging, the expression rates of GATA3 in TNM stage decreased gradually, the positive expression rates of which in stage I,II were respectively higher that in stage III(P<0.05)..However,the GATA3 expression presented no obvious difference in stage I and II.(P>0.05).In addition,GATA3 expression was not significantly associated with patient age, menopausal status and tumor size(P>0.05).2 Relationship between the expression of GATA3 and the expression of ER,PR,Her-2 and AR in breast cancerER positive expression is existing in the nucleus with a positive rate of55.5%(116/209 cases).ER positive breast cancer tissue with a positive rate of GATA3,83.6%(97/116 cases),reflects a significant difference compared to ER negative breast cancer tissues,32.3%(30/93 cases)-(P<0.05 r=0.523).There is no obvious difference(P>0.05)in the expression of GATA3 in weak, moderately expressed, strongly expressed breast cancers with a rate of 82.4%(14/17)),89.3%(25/28)and 81.7%(58/71)respectively.PR positive expression located in the nucleus, with a positive rate of 45.5%(95/209 cases). An evident difference appears in the positive rate of GATA3expressed in PR positive expression in the breast cancer tissues,82.1%(78/95 cases)and that of PR negative expressed in the breast cancer tissues,43.0%(49/114cases).Her-2 positive expression located in the cytomembrane with a positive rate of38.8%(81/209 cases). And no apparent difference exists between the positive rate of GATA3 expression breast cancer tissues,56.8%(46/81 cases)and that in Her-2negative breast cancer tissues,63.3%(81/128 cases)(P>0.05).AR positive expression located in the nucleus with a positive rate was 65.1%(136/209 cases). There is an obvious difference in the positive rate of GATA3expression in AR positive breast cancer tissue,66.9%(91/136 cases),compared to that in AR negative breast cancer tissues,49.3%(36/73 cases)(P<0.05 r=0.172).3 Relationship between GATA3 expression and Ki-67 pro-liferation index in breast cancerThe proliferation index of Ki-67 in GATA3 positive breast cancer tissues was37.51±23.40%,while that of GATA3 negative breast cancer tissues was 47.50±23.93%,with significant difference((P<0.05).The Ki-67 proliferation index of GATA3 negative breast cancer tissues obviously higher than GATA3 positive expression.4 Expression of GATA3 in Different Molecular Types of Breast CancerRatio in teiple negative and non-triple-negative breast cancer. The expression rates of GATA3 in two above preasted 17.5%(7/40 cases),and 71.0%(120/169cases)respectively,with which reflected a big difference.Ratio in Luminal type,Her-2 overexpression, and triple negative breast cancer:The expression rates of GATA3 in three types of breast cancer above presented83.6%(97/116),43.4%(23/53),and 17.5%(7/40 cases)respectively. There was an distant difference among the three groups in the expression of GATA3(χ~2=63.529,P<0.05),especially when the compansion in every two groups(P<0.05).Conclusion(1)The positive expression of GATA3 in breast cancer was related to the degree of differentiation, lymph node metastasis and TNM staging.(2)The expression of GATA3 in breast cancer tissues was positively correlated with ER,PR,AR and negatively correlated with Ki-67 proliferation index, not only suggesting that the loss of GATA3 expression may promote the proliferation of breast cancer, but also suggesting that GATA3 expression is helpful for the selection of clinical treatment options.(3)Although there were no significant difference during GATA3 expressions in breast cancer with different levels expression of ER,the GATA3-positive expression can be used as one of the auxiliary indicators of weak expression of ER.GATA3 expression detected in breast cancer tissues with weak expression of ER is conducive to clinical Endocrine therapy.(4)The expression of GATA3 in different molecular types of breast cancer has important significance and can be used as a reference standard for molecular typing of breast cancer. | | Keywords/Search Tags: | Breast
