| Objective:There are considerable individual differences in the efficacy of statins. The objective of this study was to summarize the relationship between APOE gene polymorphism and the efficacy of statins and evaluate the effect of each APOE genotype on the efficacy of statins. Methods:We first set the inclusion and exclusion criteria of literature and identified the study types and interventions.Second,we searched all the literature on statin-induced lipidlowering efficacy and APOE genotypes in PubMed,Embase,Web of Science,The Cochrane Library,Clinical Trial database. After screening the literature, the Newcastle-Ottawa quality assessment method was used to evaluate the quality of the published literature.Revman5.3 software was used for quantitative analysis after data extraction, the results were output and then explained and discussed. Results:A total of 23 articles,26 studies were included in the meta-analysis,with 6320 cases in all.The outcome measures were percentage changes of TC,LDL-C,HDL-C and TG levels. The results showed that the APOE genotypes had statistically significant difference in statininduced LDL-C reduction(E2 vs E3,MD =-2.50,P = 0.04;E4 vs E3,MD = 3.03,P <0.0001).A subgroup analysis of gender showed that differences in statin-induced blood lipids among genotype groups in male population were greater than those in females.Another subgroup analysis of the types of statin drugs showed that the efficacy of atorvastatin showing significant differences among all APOE genotypes. However, patients with FH seem to have little effect on the results. Conclusion:There was a definite difference in the reduction of LDL-C induced by statin in the APOE genotype, with a stronger difference in male and atorvastatin. |
