Objective:To investigate the expression and significance of IMP3 and S100P in mucoepidermoid carcinoma of salivary gland,pleomorphic adenoma and normal parotid gland.It may provide some advanced theoretical basis for mucoepidermoid biological targeting therapy.Methods:From 2009 to 2017,to collected Yantai Yuhuangding Hospital affiliated to Qingdao University Medical College’s 49 cases of mucoepidermoid carcinoma tissues,40 cases of pleomorphic adenoma tissue and 20 cases of normal parotid gland tissue which were confirmed by Department of Stomatology.And the expression of IMP3 and S100P protein in 49 cases of mucoepidermoid carcinoma,40 cases of pleomorphic adenoma and 10cases of normal parotid tissues were detected by immunohistochemical two-step method.Analysis of the relationship between expression of IMP3,S100P and age,sex,case classification and other related pathological features of patients with mucoepidermoid carcinoma.And the relationship between the two proteins were analyzed statistically by SPSS 19.0.Results:(1)The expression levels of IMP3 protein strongly expressed in mucoepidermoid carcinoma,but not in pleomorphic adenoma and normal parotid gland.And the expression levels of S100P protein with mucoepidermoid carcinoma tissues was higher than the pleomorphic adenoma tissues,and the pleomorphic adenoma’s expression levels was higher than the normal parotid tissues.The expression rate of IMP3 protein was46.9%,0.0%and 0.0%,while the rate of S100P protein was 59.1%,32.5%and 15.0%in mucoepidermoid carcinoma tissue,pleomorphic adenoma tissue and normal parotid tissue(P<0.01).The difference of three gruops has statistical significance.(2)The positive rate of IMP3 protein in well-differentiated salivary gland mucoepidermoid carcinoma was 32.3%and that in poorly differentiated salivary gland was 72.2%(P<0.05).The positive rate of lymph node metastasis was 78.9%,which was significantly higher than that without lymph node metastasis(26.7%,P<0.05).The expression of IMP3 protein was correlated with histological differentiation,lymph node metastasis of mucoepidermoid carcinoma(P<0.05),but no correlated with sex,age,smoking history,tumor diameter and nerve invasion.(3)The positive rate of S100P protein in well-differentiated salivary gland mucoepidermoid carcinoma was 41.9%and that in poorly differentiated salivary gland was 88.9%(P<0.05).The positive rate of lymph node metastasis was 84.2%,which was significantly higher than that without lymph node metastasis(43.3%,P<0.05).The expression of S100P protein was correlated with histological differentiation,lymph node metastasis of mucoepidermoid carcinoma(P<0.05),but no correlated with sex,age,smoking history,tumor diameter and nerve invasion.(4)The expression rate of IMP3 protein and S100P protein showed a positive correlation in mucoepidermoid carcinoma tissue(χ~2=4.740,γ=0.311).The difference of three gruops has statistical significance(P=0.029<0.05).Conclusion:(1)The expression of IMP3 in mucoepidermoid carcinoma tissues was higher than that in pleomorphic adenoma and normal parotid gland tissues;And relevant with the degree of differentiation of mucoepidermoid carcinoma and lymph node metastasis.(2)The expression of S100P in mucoepidermoid carcinoma tissues was also higher than that in pleomorphic adenoma and normal parotid gland tissues.But in pleomorphic adenoma and normal parotid gland,the expression decreased gradually;And relevant with the degree of differentiation of mucoepidermoid carcinoma and lymph node metastasis.(3)IMP3 and S100P in mucoepidermoid carcinoma tissues has positive correlation,which suggested that the synergy expression between them might be closely related to the occurrence and development of mucoepidermoid carcinoma.It may be used as a new therapeutic target for biological targeted therapy of tumor and provide a new marker for early screening,clinical diagnosis,prognosis evaluation and monitoring of mucoepidermoid carcinoma. |
