The free radicals can be produced by normal metabolism and exogenous stimuli.excessive free radicals remained may cause cell damage and apoptosis and enzyme inactivation,which leading to the aging of the body and causing a variety of diseases,such as diabetes.It was reported that collagen peptide has good antioxidant,biocompatibility and safety.It can enhance the immune and antioxidant effects of the body and is widely used in health food and biological medicine.The collagen polypeptide chromium(III)chelate has been proved to activate immune cells,improve the immune function of mice,and promote the synthesis of liver glycogen and the glucose phosphorylation in diabetic mice.Therefore,in order to better develop the medical function of collagen peptide,the paper will explore the antioxidant properties of collagen,the antioxidation of collagen peptide dissolved in different ion environment and intervention on the hypoglycemic effect of diabetic experimental animals.which will provide the basis for the in-depth study of collagen peptide.Evaluation of the antioxidative effect of collagen.With WSC,C-1,C-2,SC,SKI-ASC,WCII,WWSCII,SCII as raw materials,the relationship between the composition of collagen and its amino acid composition and antioxidant activity was analyzed by the indexes of SDS-PAGE,amino acid composition,reducing power,·OH and DPPH·Scavenging ability.SDS-PAGE analysis showed that WSC,C-1,C-2,SC and SKI-ASC were type I collagen,WCII,WWSCII and SCII were type II collagen.The content of glycine residues in collagen was 317-356 residue/1000 residues,followed by alanine residues and proline residues.The absorbance values and of reducing power,·OH and DPPH·Scavenging ability EC500 of WWSCII is 2.86,9.2 and4.3 mg/mL,respectively.the next is SCII SCII WSC WSC SKI-ASC etc.The regression equation of reductive force of WWSCII is Y=0.0241X+0.0275,the regression coefficient is above 0.99,and other collagen shows similar results.In the range of 2-10mg/mL concentration,it also showed a good dose-responserelationship.The effect of ions on the antioxidant activity of collagen peptide was judged by determining the reducing power,·OH and DPPH·Scavenging ability of collagen peptide in acetic acid,sodium acetate and calcium acetate.The results showed that the reducing power of GBB-10SP and TYPE-S decreased 0.96-2.58 fold in H+and Na+,But it has a weak effect on the scavenging activity of DPPH·.The collagen peptide had no Scavenging ability of·OH in the Ca2+.the hydroxyl radical scavenging activity was significantly increased from 11.94 to 60.56%and-4.31 to 15.90%for GBB-10SP and TYPE-S at 15 and 80 mg/mL at 0.1 M Na+which are significant difference(p<0.05),respectively.Ca2+had little effect on the reducing power,·OH and DPPH·Scavenging ability of GBB-10SP and TYPE-S.The characteristics of hypoglycemic effect of GBB-10SP were determined in mice diabetic models.The diabetic model mice were administrated GBB-10SP through intragastric administration and then the value of fasting glucose,body weight,organ mass and liver antioxidant capacity of the mice and given oral glucose tolerance test were measured.After the treatment of GBB-10SP,The area under Curve reduced to51.51-55.52 mmol/L,liver index and kidney index decreased to 40.44-41.52 mg/g,16.27-16.59 mg/g,respectively.The spleen index increased to 2.94-3.14 mg/g,Superoxide dismutase activity increased to 90.05-103.90μ/mg protein,and the content of MDA decreased to 2.94-3.03 nmol/mg protein.Compared with the model group,the AUC at 21 days decreased significantly(p<0.05),liver index and kidney index decreased,but the spleen index increased.The antioxidant ability of liver tissue increased.,but no blood sugar reduction effect,which can maintain the late blood sugar value does not continue to increase.GBB-10SP can improve of liver antioxidant capacity of diabetic mice model.Meanwhile,the treatment of collagen peptide and Momordica charantia extracts could promote organ repair and improve the ability of autoimmunity.The hypoglycemic effects of low molecular weight collagen peptide and Momordica Charantia extracts were determined in mice diabetic model in vivo.After the treatment of collagen peptide and Momordica Charantia extracts,the blood sugar value was decreased to 7.5-16.63 mmol/L,liver index and kidney index decreased to44.98-56.17 mg/g,11.29-17.95 mg/g,respectively.The spleen index increased to2.713.52 mg/g,Area under Curve reduced to 32.03-38.14 mmol/min·L.Superoxide dismutase activity increased to 89.18-154.72μ/mg protein,and the content of MDA decreased to 1.66-3 nmol/mg protein.Compared with themodel group,the bone collagen peptide of Epinephelussp and Momordica charantia extracts of the middle dosage treatment group,and the skin collagen peptide of Oreochromismossambicu and Momordica charantia extracts of middleand high dosage treatment groupseffect were remarkable.The blood sugar value at 28 days and AUC at 21 days decreased significantly,liver index and kidney index decreased,but thespleen index increased.The antioxidant ability of liver tissue increased.The results showed that the treatment of collagen peptide and Momordica charantia extracts could significantly reduce(p<0.05),and the blood glucose value related to the improvement of liver antioxidant capacity of diabetic mice model.Meanwhile,the treatment of collagen peptide and Momordica charantia extracts could promote organ repair and improve the ability of autoimmunity.Collagen has many biological activities.The results show that collagen has antioxidation,while WWSC II has significant antioxidantation.The effects of H+and Na+on GBB-10SP and TYPE-S were significantly different,while Ca2+was slightly weaker.Furthermore,GBB-10SP has no hypoglycemic effect on diabetic mice with high blood glucose level,but it can promote the repair of organs.The low molecular weight collagen peptide and Momordica Charantiaextracts significantly reduced the blood sugar of diabetic mice(p<0.05),and the blood glucose value related to the improvement of liver antioxidant capacity.Meanwhile,the treatment of collagen peptide and Momordica charantia extracts could promote organ repair and improve the ability of autoimmunity.
