Evaluation Of HSP60 On The Diagnosis In Colorectal Cancer By ROC

Objectives The serum concentrations of heat shock protein 60(HSP60)in patients with colorectal cancer were detected for the analysis of their relationgship.The evaluation on the diagnosis of HSP60 linked with the other 4 tumor markers in patients with CRC was performed,so to offer an experimental basis for the screen and specific diagnosis of CRC.Methods 156 cases of colorectal cancer in Tangshan City people’s Hospital from June 2017 to December 2017 were selected(all cases were selected by pathological examination),of which 102 cases were male,age 61.0±10.0 years,54 women,60.6±10.6 years old as colorectal cancer group,and another 40 healthy physical examiner in the Tangshan City people’s Hospital in the same period were selected as normal control group,of which 25 were males,45.1±10.3 years old,and 15 women,47.1±7.8 years old.The serum levels of HSP60 were measured by ELISA method and the contents of CEA,CA242 and CA724 in serum were measured by electrochemiluminescence.The content of serum CYFRA21-1 was determined by chemiluminescence immunoassay.The correlation between HSP60,CEA,CA242,CA724,CYFRA21-1 and colorectal cancer was analyzed.On this basis,ROC were adopted to evaluate the diagnostic value of combined detection of HSP60 linked with CEA,CA242,CA724 and CYFRA21-1 in colorectal cancer.Results 1 The contents of HSP60 in CRC group and normal control group were 481.79±417.52pg/mL and 184.85±92.16pg/mL respectively,and the difference was statistic significant(P<0.001).The contents of HSP60 in stage I+II CRC patients and stage III+IV ones were 383.90±187.84pg/mL and 747.49±680.31pg/mL respectively,and the difference was statistic significant(P<0.001).The serum concentration of HSP60 was irrelevant to sex,age and pathologice location in patients with CRC,and the difference was not statistic significant(P>0.05).2 The serum concentrations of CEA,CA242,CA724 and CYFRA21-1 in CRC were 31.38±95.13ng/mL,20.42±36.06U/mL,6.35±5.12U/mL and 5.12±4.18ng/mL respectively,while they were 2.12±1.52ng/mL,3.68±1.93U/m L,1.57±1.32U/m L,1.98±0.88ng/mL respectively in normal control group,and all of differences about the above items were statistic significant(P<0.01).3 The contents of CEA,CA242,CA724 and CYFRA21-1 in stsge I+II were 10.18±22.67ng/m L,12.67±25.57U/mL,3.47±4.39U/mL and 8.63±3.95ng/mL respectively,while they were 88.90±167.79ng/mL,41.48±49.90U/m L,14.19±17.29U/m L and 10.08±4.98ng/mL respectively in stage III+IV,and the differences of CEA,CA242 and CA724 between stage I+II and stage III+IV were statistic significant(P<0.01).while the difference of CYFRA21-1 between stage I+II and stage III+IV was not statistic significant(P>0.05).4 The AUC of HSP60,CEA,CA242,CA724 and CYFRA21-1 were 0.858,0.801,0.706,0.708 and 0.793 respectivel y,Cut-off values are 165.41pg/m L,1.67ng/mL,5.29U/m L,0.64U/mL,and 2.48ng/mL respectively,the sensitivities were 98.7%,85.9%,51.3%,84.6% and 73.1% respectively,the specificities were 55.0%,62.5%,95.0%,40.0% and 87.5% respectively.The Youden index were 0.537,0.484,0.463,0.381 and 0.606 respectively,5 The AUC of HSP60 joint with CEA,CA242,CA724 and CYFRA21-1 in the diagnosis of CRC were 0.924,0.896,0.903 and 0.918 respectively,and the sensitiviti es for the screen of CRC were 99.82%,99.37%,99.80% and 99.66% respectively.6 The AUC of HSP60 serieses with CEA,CA242,CA724 and CYFRA21-1 in the diagnosis of CRC were 0.904,0.926,0.897 and 0.920 respectively,and the sp ecificities for the screen of CRC were 83.13%,97.75%,73.00% and 94.38% respe ctively.Conclusion 1 The serum content of HSP60 was revelant to CRC and it’s pathologic TNM.2 The serum concentrations of CEA,CA242,CA724 and CYFRA21-1 were revelant to CRC and it pathologic TNM,but CYFRA21-1 was only revelant to CRC.3 The diagnosis effectiveness of HSP60 and CEA were higher than the other 3 umor markers in the diagnosis of CRC,and that of CYFRA21-1 was high,while those of CA242 and CA724 were medium in the diagnosis of CRC.4 HSP60 joint with CEA might be adopted to the screen diagnosis of CRC,while HSP60 serised with CA242 was suitable for the specific diagnosis of CRC.
