Coptis Chinensis(CC)is widely used to treat diabetes in Chinese medicine for its significant hypoglycemic and hypolipidermic effect.In this study,we established a rat model of type 2 diabetes and then treated with supercritical-extracted CC and gastric juice-extracted CC,respectively.Body weight,oral glucose tolerance test(OGTT),fasting plasma insulin level and lipid profile were measured and insulin resistance index was calculated.Plasma proteins between type 2 diabetic rats and CC-treated rats were identified using an iTRAQ-based quantitative proteomic analysis.1.5 times of the protein was analyzed by bioinformatics.The results showed that the plasma levels of total cholesterol(TC),triglyceride(TG)and low density lipoprotein(LDL)in rats of both CC-treated groups were significantly decreased.Proteomics identified 929 proteins,and 15 proteins were selected based on their expression levels and bioinformatics.Among these proteins,9 proteins(IGF-1,IGFBP-4,IGFBP-6,Igfals,C2,C4,Cfi,Prdx-2 and Prdx-3)were up-regulated in two CC-treated groups while 6 proteins(Pla2g7,Pcyoxl,ApoC-1,ApoC-3,ApoB-100 and ApoE)were down-regulated.Their function is associated with glucose mebobolism,insulin action,immunity,inflammation,lipid metabolism,oxidation and antioxidation.Two differently extracted CC treatments did not show significant differences in the results.The research reinforces our understanding of the therapeutic effects and mechanisms of CC in the treatment of type 2 diabetes.
