Aims:This article explored the expression of Rab1 A gene in colorectal cancer.Through regulating the expression of Rab1 A gene,the effect of regulating Rab1 A expression on the ability of proliferation,migration and invasion in colon cancer cell lines was investigated,which provides the basis for Rab1 A to be a vital target for individualized treatment of colorectal carcinoma.Methods:The expression of Rab1 A in 100 colorectal cancer tissues and adjacent normal tissues was detected by immunohistochemistry(IHC).Real-time quantitative PCR was used to investigate the m RNA expression of Rab1 A in 15 colorectal tissues(including normal tissue,adenoma and adenocarcinoma,respectively,in five cases).we researched Oncomine to analysis the expression of Rab1 A gene in cancer of the colon,colorectal mucosal carcinoma and normal colorectal tissue.Western blotting(WB)was used to detect the expression of Rab1 A protein in six kinds of common colorectal carcinoma cell lines.Screening Rab1A-highly expressed cell lines,then targeted knockdown of Rab1 A high-expressing cell lines by si-RNA.The MTT methods,Transwell assays and wound healing were used to evaluate the cell’s proliferation,invasion and migration capabilities.Results:Detecting the expression of Rab1 A in 100 cases of colorectal cancer and 29 cases of adjacent tissues by immunohistochemistry technique,we found that the expression of Rab1 A genes in colorectal tissue significantly higher than in normal tissues(adjacent tissues).The positive rates of Rab1 A in colorectal cancer and adjacent tissues were 78% and 24%,respectively,obvious difference was statistically significant(p=0.001).Subsequently we researched Oncomine to show that Rab1 A expression was significantly higher in colorectal cancer than in normal tissues.Then subgroup analysis revealed that Rab1 A expression in colon mucosal carcinoma and the rectal mucosal carcinoma was apparently higher than that in normal colon tissues.The differences were statistically significant,and the p value was 0.032 and 0.015 respectively.Quantitative PCR was used to measure the m RNA expression of Rab1 A in 15 colorectal specimens(five cases of normal tissue,adenoma,and adenocarcinoma).Discovered that the expression of Rab1 A in colorectal cancer tissues was distinctly higher than that in adjacent normal tissues and adenomas,but there was no significant difference between adjacent tissues and adenomas.In addition,SW480 was highly expressed by Western blotting,and the ability of proliferation and migration of SW480 cells was found to be closely related with high expression,and the proliferation,migration and invasion ability of SW480 cells could be significantly inhibited by targeting and down regulating Rab1 A expression.Conclusions:Rab1A is an oncogene of colorectal cancer.Targeting down-regulation of Rab1 A gene expression obviously inhibites the proliferation,migration,and invasion of colon cancer cell lines.Rab1 A is expected to be a new biomolecule in individualized therapy as a new target in the early diagnosis of colorectal cancer. |
