Effect And Mechanism Of MiR-1225-5p Regulating Proliferation,Migration And Invasion In Colorectal Cancer Cells | Posted on:2023-10-01 | Degree:Master | Type:Thesis | Country:China | Candidate:Z H Zhang | Full Text:PDF | GTID:2544307031457364 | Subject:Surgery | Abstract/Summary: | PDF Full Text Request | Objectives To construct a model of miRNA-1225-5P analog and explore the effects of miRNA-1225-5P on LOVO proliferation,migration and invasion of human colorectal cancer cells,so as to provide partial data and experimental reference for the study of miRNA-1225-5P in human colorectal cancer cells,as well as for the diagnosis and prognosis of human colorectal cancer.Methods Human colon cancer cell lines HCT116,HT-29 and LOVO were cultured.The expression level of miRNA-1225-5P in each group of colorectal cancer cells was detected by Q-PCR,and the cell line with the greatest difference was screened out.The screened colorectal cancer cell line LOVO was cultured and transfected with miRNA-1225-5P MIMics and meaningless sequence NC.The experiment was divided into blank group,experimental group and NC group.The expression level of miRNA-1225-5P in the three groups of colorectal cancer cells was detected by Q-PCR,respectively,to verify whether the transfection was successful.After successful transfection,the increment ability of colorectal cancer cells was detected by CCK-8 method,and the migration and invasion ability of colorectal cancer cells were detected by scratch test and Transwell test.Western blot was used to detect the expression levels of colorectal cancer related antibody proteins Vimentin,N-cadherin and E-cadherin.Results The expression levels of miRNA-1225-5P in human colorectal cancer cells HCT116,HT-29 and LOVO were all low,and the relative expression level of Lo Vo in colorectal cancer cells was the lowest.Q-PCR results showed that miRNA-1225-5P mimics significantly up-regulated the expression level of miRNA-1225-5P in colorectal cancer cells LOVO.CCK-8 assay showed that up-regulation of miRNA-1225-5P could enhance the proliferation of LOVO in colorectal cancer cells.Scratch assay and Transwell assay showed that up-regulation of miRNA-1225-5P decreased the migration and invasion ability of colorectal cancer cells LOVO.Western Blot results showed that the expression of N-cadherin protein in LOVO of colorectal cancer cells was increased after up-regulated miRNA-1225-5P expression.Conclusions Overexpression of miRNA-1225-5P can promote the proliferation of colorectal cancer cells to a certain extent,inhibit the migration and invasion of colorectal cancer cells,and increase the expression of related proteins.Figure 7;Table 15;Reference 101... | Keywords/Search Tags: | Colorectal cancer, The miRNA-1225-5p, Cell proliferation, Cell migration, Cell invasion | PDF Full Text Request | Related items |
