| Benign prostatic hyperplasia(BPH)is a disease in medium-elderly men.it causes urgency,urinary frequency,dysuria and a series of urinary tract symptoms because of cell hyperplasia in prostate tissue.More and more people are affected by this disease,and it is very important to improve the quality of life in medium-elderly men.The imbalance of sex hormone level is the key point of this disease.The treatment of BPH is mainly based on western medicine and surgery.Chinese traditional medicine such as acupuncture and massage can improve the BPH by regulate yin and yang.Acupuncture has the advantages of simple and non-toxic effect,and the clinical effect is remarkable.But there are few studies on the mechanism of acupuncture on BPH.The current research on the mechanism of BPH is broadly divided into two aspects: one is the relationship between the hormone level and the growth factor,and the other is the apoptosis function of the cell.The role of acupuncture in treating BPH and regulating the level of sex hormone to influence the relationship between prostate cells need to be further explored.Therefore,this research discussed sex hormone levels to provide more scientific basis for acupuncture treatment of BPH by Jin’s-3 needle therapy "three needles of urine".ObjectiveThe main points for the treatment of urinary system diseases in Jin’s-3 needle therapy are "three needles of urine"(Guan Yuan,ZhongJi and San Yin jiao).Clinically,it is effective to treat BPH with a combination of syndrome differentiation and acupuncture.Therefore,in this experiment,the prostate index was calculated by studying three needles of urine in the BPH model.Detect the serum DHT,estradiol and related growth factor,basic fibroblast growth factor and transforming growth factor beta 2 expression,the influence of the clear three needles of urine for treatment of BPH effect and the mechanism.Provide scientific theory and basis in the treatment of BPH for clinical use of three needles of urine.MethodsIn this study,18 male SD rats were randomly divided into 3 groups: normal group,model group,and acupuncture group,6 rats in each group.To be closer to the reality,the model group and acupuncture group were injected of testosterone propionate for 28 days to establish model of BPH.Normal subcutaneous injection group were given the corresponding amount of olive oil at the same time.From the 29 th day,the acupuncture group was engaged in acupuncture intervention,while the model group and the normal group were engaged for 21 consecutive days at the same time.After the last intervention,all rats were fasting for 12 hours and water was forbidden for 6 hours,and the abdominal aorta was taken after weighing.The expression of serum dihydrotestosterone,estradiol,alkaline fibroblast growth factor,and transformation growth factor of the rat serum was determined by the enzyme linked immunoassay(ELISA).The complete prostate tissue was taken out and weighed to calculate the prostate index(PI).Results1.Calculation results of the prostate index(PI)Compared with the normal group,the prostate index of the model group and the acupuncture group was significantly increased(P < 0.01).Compared with the model group,the prostate index of acupuncture group decreased significantly(P < 0.01).2.ELISA test results of enzyme-linked immunosorbent assay(ELISA):(1)dihydrotestosterone: the expression rate of DHT in the acupuncture group was lower than that in the model group(P < 0.05).The expression rate of model group was significantly higher than that in normal group(P < 0.01).There was no statistical difference between the groups(P >0.05).(2)estradiol: the estradiol expression rate of the acupuncture group was lower than that in the model group,but there was no statistical difference(P > 0.05).The expression rate of the model group was higher than that in the normal group(P < 0.05).There was no statistical difference between the groups(P > 0.05).(3)basic fibroblast growth factor: the expression rate of basic fibroblast growth factor of acupuncture group was significantly lower than that in the model group(P < 0.01).The expression rate of model group was significantly higher than that in normal group(P < 0.01).There was no statistical difference between the groups(P >0.05).(4)transformation growth factor β 2: the expression rate of the transformation growth factor β 2 in the acupuncture group was lower than that in the model group,but there was no statistical difference(P >0.05).The expression rate of the model group was higher than that in the normal group(P < 0.05).There was no statistical difference between the groups(P >0.05).ConclusionIn the study,the serum dihydrotestosterone and growth factor basic fibroblast growth factor expression in serum of BPH rats were decreased after three needles of urine.But the effect on estradiol and transforming growth factor β 2 was not obvious.It can be concluded that the treatment of BPH with three needles of urine is mainly by reducing the expression of dihydrotestosterone and its related basic fibroblast growth factor to reduce the hyperplasia of prostate cells and improve a series of urinary system symptoms.It could provide basic theoretical support for the clinical use of acupuncture therapy for BPH. |
