| Background: according to the international cancer research(IARC),pulmonary malignancies still take the leading place of the global malignancy and are the cause of the second common female disease in the world.According to the Chinese who study China’s cause of death,for the third time in recent 30 years,China’s lung cancer mortality rate increased by 46.5%,especially the past 20 years China’s rapid rise,the prevalence of female lung adenocarcinoma is twenty-one over one hundred thousand.It has been investigate that male sex is much higher in most country,and that ratio is between 2 and 16:1,but the ratio of the sex ratio in China is close to 1:1.As is known to all,smoking is generally recognized as having a close relationship with lung cancer.However,in terms of active smoking rates,Asian women are generally much lower than men and women in Europe and the United States.In particular,there is no increase in smoking and frequency among Chinese women,but the prevalence rate is rising rapidly.It show that there must be a close link between that causes of smoke and lung adenocarcinoma in women.Foreign scholars have also found that the three factors of non-smoking,Asian women and lung adenocarcinoma tend to be closely related,that is to say,Asian non-smokers are characterized by histological characteristics: most patients with lung adenocarcinoma are women.Women suffer from lung cancer mostly adenocarcinoma.If this type of female tumor is isolated as a unique subgroup analysis,such patients usually do not have any unsatisfactory clinical symptoms in the early stage,and they can only maintain a short period of survival with a short survival period,and miss the best cure.Therefore,early diagnosis and treatment of such patients is crucial.At present,however,the early diagnosis and screening of such patients is not ideal,and still mainly depends on image and sputum examination,and there is a certain omission and misdiagnosis,though the pathological examination and diagnosis rate is high,but the wounds are relatively large.In recent years,metabolomics has made great progress in the early diagnosis,prognosis and pathogenesis of lung adenocarcinoma of Chinese women on the research object,through the metabonomics method to explore its metabolites the main differences between the biological function and metabolic mechanism,and find the potential diagnostic markers,which has important clinical value and significance in the aspect of prevention,early diagnosis and individualization treatment of these patients.Objective: 1.Based on metabonomics,targeted analysis in nonsmoking women in patients with lung adenocarcinoma,non smoking women’s health and nonsmoking women patients with lung benign tumor metabolic substances in the serum were analyzed,and observe the metabolic profile changes,well differentiate between three different groups,to find potential diagnostic markers,thus to women for clinical screening and diagnosis of lung adenocarcinoma.2.It collect clinical data,and detects that change of different metabolite in serum of patients with such adenocarcinoma,and then explores its biological function and metabolism mechanism.Methods: to collect 65 non-smoking female lung adenocarcinoma patients,65 cases of non smoking women’s health,and 6 cases of non smoking women lung benign tumors patients plasma and remove protein in plasma and serum after frozen keeping.Sample pretreatment before experiment,chromatography,mass spectrometry detection,data analysis,etc.Results: 1.65 cases of non-smoking female lung adenocarcinoma were significantly distinguished from 65 non-smoking healthy women,and 45 significant differences were found.The results showed that compared with normal women,non-smoking female lung adenocarcinoma patients had different degrees of disorder in energy metabolism,amino acid metabolism,oxidative stress and lipid metabolism.2.Through ROC curve analysis and external validation,a group of composite markers composed of cysteine,serine and monacyl glyceride were selected as diagnostic markers for the potential lung adenocarcinoma of non-smoking female patients.Conclusion: 1.The experiment using metabonomics method,65 cases of non smoking women patients with lung adenocarcinoma and 65 cases of healthy women smoking serum sample data were analyzed,and successfully distinguish the lungadenocarcinoma patients and healthy people,using multivariate and univariate analysis,we successfully get 45 metabolite changes significantly,the results showed that the metabolite changes,compared with healthy,nonsmoking women patients with lung adenocarcinoma of energy metabolism,amino acid metabolism,oxidative stress andlipid metabolism were happened to a certain degree of disorder.2.Through external validation strategy,we successfully screened by serine,cysteine,single acyl glyceride combined into groups and markers as nonsmoking women diagnostic tumor markers in patients with lung adenocarcinoma. |
