The induced pluripotent stem cells(iPSCs)reprograms somatic cells to exhibit a pluripotent stem cell-like state similar to that of embryonic stem cells.This revolutionary technology won Yamanaka Nobel Prize in Physiology and Medicine in 2012.Edwards syndrome,also known as trisomy 18,is the second most common autosomal syndrome.Chromosomal abnormalities are due to the presence of three chromosomes in patient 18.The main symptoms of Edwards syndrome are a variety of organ malformations,including abnormalities in the heart and kidneys,characteristic craniofacial features et.al.The infant mortality rate is high,and the surviving patients all have severe growth retardation.The main causes of death include central apnea,heart failure caused by heart failure,and respiratory diseases.We used human skin cells derived from skin and amniotic fluid from hospitals,and conducted primary isolation and culture of fibroblasts and amniotic fluid cells.Non-integrated plasmids were electroporated and reprogrammed to induce pluripotent stem cells.After purification,their pluripotency was identified,including alkaline phosphatase enzyme staining,rt-PCR detection,embryoid body formation,and flow cytometry detection.In the process of exploring iPSCs production,we established models of Edwards symptoms and congenital inversion of iPS cells,contributing to rare laboratory research.Generation of induced pluripotent stem cells offering a method for research genetic disease.We obtain may be genes associated with trisomy 18 malformations by RNA-Sequencing analysis technology.
