| Objective: By observing the effects "Dao Di Tong Guan Tang" on the change of the oviduct inflammatory tubal histomorphology in rats and tubal tissues smad2,smad7 protein and the changes of ultrastructure of mucosa molecules expression effect,discussing "Dao Di Tong Guan Tang" explore smad2 and smad7 relation with the pathogenesis of salpingitis,further defined authentic "Dao Di Tong Guan Tang" anti-inflammatory and anti aging mechanism,thus provided the scientific basis for clinical treatment of oviduct inflammatory obstructive disease.Methods :Eighty female SD rats by method of sampling random list 12 for the normal group,the remaining 68 vaccinate "bacteria suspension liquid" to make model rats with chronic tubal phlogistic sex,respectively to observe in the mould made model rats in 5,10,15,20 days after the fallopian tube,the morphology and pathological observation,to confirm the building successfully.Building after 20 days(excluding 3 death,5 sampled)only remaining 60 rats,according to the method of random arrangement table are divided into: model group,the Kang Fu anti-inflammatory positive control group,"Dao Di Tong Guan Tang" low,medium and high dose group,each group of 12.Raised in the same conditions,starting building 20 days after enema,which the conventional breeding normal group and model group,positive control group to Kang Fu antiphlogistic suppository water solvent,"Dao Di Tong Guan Tang" low,medium and high dose groups respectively to authentic flux "Dao Di Tong Guan Tang" low,medium and high concentration of water decoction.Each group is divided into two periods,respectively in the enema,based in 15 days and 30 days by HE staining observation oviduct tissue morphology andimmunohistochemical technology testing oviduct tissue Smad2,Smad7 protein expression,q RT-PCR detection Smad7 m RNA content changes,scanning electron microscopy(sem)to observe the changes of the ultrastructure of tubal mucosa of rats.Results:(1)Approved by the drugs enema treatment for 15 days and 30 days after the tissue morphology and pathological observation rat oviduct,fallopian tube of model rats was found in each treatment group were have different degrees of improvement,with "Dao Di Tong Guan Tang" high dose group of the most prominent.(2)In drug delivery of 15 days and 30 days,observing each group tubal tissues of rats Smad2 Smad7 protein expression in:A.Compared with normal group,model group of Smad2 content increased,the content of Smad7 decreased significantly,there are significant difference(P < 0.05).Compared with model group,the expression of Smad2 quantity of the treatment group are reduced,Smad7 expression quantity increased,the "Dao Di Tong Guan Tang" the expression of Smad2 significantly lower than the model group,expression of Smad7 significantly higher than the model group,with significant difference(P < 0.05).For 15 days and 30 days,the treatment group dosing 30 days is different than the number of drug delivery15 days Smad2 expression degree reducing,Smad7 expression quantity increased to varying degrees,statistically significant difference(P < 0.05).(3)the content change of Smad7 m RNA: compared with the normal group,the expression quantity of the model group decreased significantly,and the difference was statistically significant(P<0.05).Than with the model group,kang fu anti-inflammatory bolt group and "Dao Di Tong Guan Tang" each expression capacity increased to different extent,among them,15 days,high "Dao Di Tong Guan Tang" group has statistical significance(P < 0.05),while for 30 days,the "Dao Di Tong Guan Tang" each group were statistically significant(P < 0.05).Compared with each group of the "Dao Di Tong Guan Tang",the content of 30 days was higher than that of 15 days,which was not statistically significant(P > 0.05).(4)for 15,30 days to observe each group the tubal mucosa ultrastructure changes in rats: normal group rats is widely populated cilia,tubal mucosa surface model groups see extensive cilia fall off,ciliated cells decreased significantly and the extensive adhesion,shorter cilia,sparse distribution and irregular.The condition of oviduct cilia was improved with different degrees of the kangfu anti-inflammatory suppository group and each group of the "Dao Di Tong Guan Tang",.Conclusion :(1)Through the immunohistochemical detection and q RT-PCR detection technology,can be found when inflammation is aggravating,oviduct tissue in rat models of Smad2 expression quantity increases,the expression of Smad7 volume decreases,and when the inflammation control Smad2 expression quantity after cut,the expression of Smad7 quantity to increase,this may explain Smad2,Smad7 is involved in the establishment of the oviduct inflammatory process,"Dao Di Tong Guan Tang" soup can effectively inhibit Smad2 expression,promote the Smad7 protein expression level,"Dao Di Tong Guan Tang" soup of anti-inflammatory and anti fibrosis mechanism maybe related to inhibit Smad2 and promote Smad7.(2)After drugs enema treatment,by the naked eye and biopsy pathology,each rat oviduct organization of scanning electron microscopy(sem)observation,"Dao Di Tong Guan Tang" group can improve the color,of the rat oviduct tissue edema,flexibility and surrounding tissue adhesion,and improve the organization of inflammatory cells infiltration and fibrosis,improve and repair the tubal mucosa structure,to restore the fallopian tube transport function.Moreover,the longer the drug administration time,the better the effect,and the most prominent in the high group of "Dao Di Tong Guan Tang". |
