| The Eomecon chionantha Hance(E.Chionanthais)is a herbaceous perennid,which belongs to Papaveraceae,Chelidonium,Eomecon.As traditional medicine,it’s bitter and cold with little poison,and fond of shade.The plant is rich in alkaloids,and has many pharmacological activities.The purpose of this study is to find the effective active ingredients,separate the chemical constituents and determinate the structures and the antioxidant activity.1.Through soaking,the petroleum ether extraction was got as non polar part of E.chionantha.In order to research the chemical constituents of the plant roundly,the non polar part was detected by gas chromatography-mass spectrometry(GC-MS).The results of GC-MS analysis showed that the content of low boiling point was low,the relative content of lipid was as high as 82.15%,the relative content of hydrocarbons was 4.81%,ether 1.31%,ketone 7.54%,the low boiling point part contains(Z,Z)-9,12-diene-eighteen carbon acid methyl ester,(Z)-9-sixteen carbon acid methyl ester and 1-ene sixteen carbon acid.2.The total alkaloids were extracted through acid extraction and alkali precipitation.Six compounds were obtainedfrom total alkaloids by silica gel col-umn chromatography,LH-20 chromatography,recrystallization.And through1H-NMR,13C-NMR,IR,UV,X-RAY,DEPT and HMBC,six compounds were identified,which included four benzophenanthridine class alkaloids:6-keto-dihydrocannabinine(1),(l′.E)-6-(3′′-Methoxy-4′′-hydroxyphenyl)ethenyldihydrosanguinarine(2),Bocconoline(3),8-hydroxy-dihydrocannabinine(4),and 2 protopine alkaloids:Protopine(5)and allocryptopine(6).3.The antioxidant activities of 2 alkaloids in vitro were discussed by determining the total antioxidant capacity of protopine and allocryptopine(FRAP),free radical scavenging capacity of DPPH,and inhibition rate of adjacent benzene three phenolic autoxidation.The results show that the antioxidant capacity of protopine was 0.1 mg/m L,total antioxidant activity was 0.096 mmol/LFe SO4,allocryptopine antioxidant capacity at 0.1 mg/m L,the total antioxidant activity was 0.080mmol/LFe SO4;The clearance ability of DPPH scavenging activity was 1.616 and0.520,respectively.The scavenging rate of superoxide radical was up to 29.7%(1mg/m L),with a total of 14.1%(1 mg/m L).This result showed a certain dose effect relationship. |
