| Objective: by detecting myocardial infarction rats serum CD34 +,CD133 +,vascular endothelial growth factor(VEGF)and nitric oxide(NO)content,explore Zhi-Long Huoxue)capsule on myocardial infarction in the rat bone marrow endothelial progenitor cell mobilization effects and possible mechanism.Methods: healthy SD rats 48,were randomly divided into control group,captopril group,model group,Zhi-Long Huoxue capsule high-dose group)(hereinafter referred to as Zhi-Long high-dose group),Zhi-Long Huoxue capsule dose group)(hereinafter referred to as Zhi-Long dose group),Zhi-Long Huoxue capsule low-dose group)(hereinafter referred to as Zhi-Long low dose group),each group of eight.Coronary artery ligation of copying the rat model of acute myocardial infarction(AMI).Control line open thoracic surgery,broke the pericardium and exposed to the heart,not ligation of coronary artery.Record electrocardiogram(ECG)and 2 hour after observing changes of ST segment and waveform.Postoperative 1 day,Zhi-Long the high dose group,dose group,Zhi-Long low dose group,respectively,to be 1.6 g/kg/d,0.8 g/kg/d,0.4 g/kg/d doses of Zhi-Long Huoxue)capsule(hereinafter referred to as"Zhi-Long)suspension liquid lavage 3 weeks;Captopril group to be 20mg/kg/d amount of captopril suspension liquid lavage 3 weeks;Control group,model group were amount of saline lavage for three weeks.Groups of rats in postoperative day 0,1 day,3 days,5 days,7 days,20 days caudal vein blood sampling method in venous blood 0.5 1 ml,flow cytometry instrument in serum was detected in the content of CD34 +,CD133 +;Gastric vein in venous blood back 21 days 2 ml,ELASA method to detect the serum level of VEGF,nitrate reductive enzymatic detection in serum NO level;Put to death in the rat,the heart blood directly for 3 to 5 ml,isolate mononuclear cells(monocyte newsun focus)layer,cultivating four days,observe the endothelial progenitor cells.Results:(1)In serum of CD34 + content: compared with control group,each observation point model group,captopril group,Zhi-Long high dose group,Zhi-long dose group,Zhi-Long low dose group serum content of CD34 + were higher,the difference is statistically significant(P<0.001);Compared with model group,the observation point the captopril group,Zhi-Long high dose group,Zhi-long dose group,Zhi-Long low dose group CD34 + were higher,statistically significant difference(P<0.001);In comparison with the captoppuri group,there was no significant change in the CD34 + content in the Zhi-Long high dose group serum of the perveron,and the difference was not statistically significant(P>0.05).In comparison with the Zhi-long dose group,the CD34 +content of the ctopuri group and Zhi-Long high dose group serum of vermiculosaurus were elevated,and the difference was statistically significant(P<0.001).In comparison with Zhi-Long low dose group,the CD34 + content of the ctopuri group and Zhi-Long high dose group serum of vermiculosaurus were elevated,and the difference was statistically significant(P<0.001).(2)In serum of CD133 + content: compared with control group,each observation point model group,captopril group,Zhi-Long high dose group,Zhi-long dose group,Zhi-Long low dose group serum content of CD133 + were higher,the difference is statistically significant(P<0.001);Compared with model group,the observation point the captopril group,Zhi-Long high dose group,Zhi-long dose group,Zhi-Long low dose group CD133 + were higher,statistically significant difference(P<0.001);In comparison with the captoppuri group,there was no significant change in the CD133 + content in the Zhi-Long high dose group serum of the perveron,and the difference was not statistically significant(P>0.05).In comparison with the Zhi-long dose group,the CD133 +content of the ctopuri group and Zhi-Long high dose group serum of vermiculosaurus were elevated,and the difference was statistically significant(P<0.001).In comparison with Zhi-Long low dose group,the CD133 + content of the ctopuri group and Zhi-Long high dose group serum of vermiculosaurus were elevated,and the difference was statistically significant(P<0.001).(3)Serum VEGF concentration: compared with control group,model group,captopril group,Zhi-Long high dose group,Zhi-long dose group,Zhi-Long low dose group serum VEGF content were elevated,statistically significant difference(P<0.001);Compared with model group,captopril group,Zhi-Long high dose group,Zhi-long dose group,Zhi-Long low dose group VEGF content were elevated,statistically significant difference(P<0.001);There was no significant change in the amount of VEGF content in the high-dose serum of vermiculosaurus,compared with the captopril group,and the difference was not statistically significant(P>0.05).The levels of VEGF in the captoppuri and Zhi-Long high dose group were higher in the group compared with Zhi-long dose group,and the difference was statistically significant(P<0.001).Compared with Zhi-Long low dose group,captopril group,Zhi-Long high dose group of serum VEGF levels were elevated,statistically significant difference(P<0.001).(4)Serum NO concentration: compared with control group,model group,captopril group,Zhi-Long high dose group,Zhi-long dose group,Zhi-Long low dose group serum NO content were elevated,statistically significant difference(P<0.001);Compared with model group,captopril group,Zhi-Long high dose group,Zhi-long dose group,Zhi-Long low dose group NO content were elevated,statistically significant difference(P<0.001);There was no significant change in the amount of NO content in the high-dose serum of vermiculosaurus,compared with the captopril group,and the difference was not statistically significant(P>0.05).The levels of NO in the captoppuri and Zhi-Long high dose group were higher in the group compared with Zhi-long dose group,and the difference was statistically significant(P<0.001).Compared with Zhi-Long low dose group,captopril group,Zhi-Long high dose group of serum NO levels were elevated,statistically significant difference(P<0.001).Conclusion: 1.The acute myocardial infarction(mi)is one of the induced factors of EPCs mobilization of rat bone marrow,Zhi-Long Huoxue)capsule can effectively promote the rat bone marrow mobilization EPCs from peripheral blood after myocardial infarction.2.Zhi-Long Huoxue Tongyu capsule,which increases the level of VEGF by increasing the amount of NO in the serum,promotes the effect of the bone marrow EPCs mobilization.3.Zhi-Long Huoxue)capsule on myocardial infarction in the rat bone marrow endothelial progenitor cell mobilization effect has certain concentration-response relationship. |
