| Objective:To establish the animal model of golden hamster of aspirin resistance (AR).To estimate the intervention of Zhilong Huoxue Tongyu capsule to the animal model of AR,and investigate the possible mechanism.Methods:Divided the 130 golden hamster into two groups randomly,10 rats in control group and 120 rats in model group.To establish the model of AR,and give model group the diet.of rich of fat sugar and salt 12 weeks, and gastric irrigation of drugs (0.74g/Kg/d of Aspirin+2.22g/Kg/d of IbuprofenTablets) and passive smoking 4 weeks.According the platelet aggregation test to be the standard to determine the AR.As follows that using Adenosine diphosphate (ADP) as a inducer to do the Platelet aggregation test and then the mean platelet aggregation≥70%,using arachidonic acid(AA) as a inducer to do the test and the result≥20%.who both meets the standards are AR,complying with one of the standards are aspirin semi-resistance(ASR).Divided into three groups of 10 rats each from AR or ASR animal models,as follows that model group,the treatment group of clopidogrel and the treatment group of Zhilonghuoxuetongyu capsule.Then giving their gastric irrigation of drugs once a day for 4 weeks.At last do the platelet aggregation test and the enzyme-linked immunoassay (ELISA) of thromboxane B2(TXB2) and 6-keto-prostaglandinF1a (6-keto-PGF1a),taking stomaches into 10% formaldehyde for the pathology testing.Results:1.The platelet aggregation(PAG) of AR and ASR rats were higher than control group's levels significantly(P<0.01).There had 6 rats were AR,and its incidence rates were 5%.There had 28 rats were ASR,and their rates were 23%.2.To compare the PAG of drug intervention before and after,the treatment group of Zhilonghuoxuetongyu were both dropped (P<0.05),and the treatment group of clopidogrel dropped in PAG of ADP-induced(PAGadp) (P<0.05).To compare Among four groups after the intervention,there were no significant differences between the treatment group of clopidogrel and the treatment group of Zhilonghuoxietongyu in PAGadp and PAG of AA-induced (PAGaa).The two groups were lower than model group in PAGadp and PAGaa (P<0.05,0.01).Compare to control group,there were significant differences with model group, the treatment group of clopidogrel and the treatment group of Zhilonghuoxietongyu both in PAGadp and PAGaa(P<0.01).3.Compared to TXB2,6-K-PGF1a and TXB2/6-K-PGF1a among four groups,both the treatment group of clopidogrel and the treatment group of Zhilonghuoxietongyu,they were lower than model group in TXB2 and TXB2/6-K-PGF1a.And the treatment group of Zhilonghuoxuetongyu were higher than the treatment group of clopidogrel and model group in 6-K-PGF1a (P<0.05).There were no significant differences when clopidogrel group were higher than model group (P> 0.05).4. The Analysis of Correlation, TXB2 were positively correlated with PAGadp, PAGaa(r=0.934,0.811,P<0.01).And the same as TXB2/6-K-PGF1a and PAGadp, PAGaa(r=0.874,0.724,P<0.01,0.05).While there were no binds between 6-K-PGF1a and PAGadp,PAGaa(r=-0.603,-0.454,P>0.05).5.The results of pathology testing of gastric,there were part sheddings of epithelial and little infiltrations with inflammatory cells in mucosa in modle group.They were serious than modle group's pathological changes in huoxuetongyu group.And there had ulcers in clopidogrel group.Conclusion:It was successful with making AR animal models,because of simulation of pathological environments and comprehensive role of multiple risk factors.Zhilong Huoxue Tongyu capsule could decrease platelet aggregation effectively,and improved AR effectively.Also because of it could reduced TXB2 and increased 6-K-PGF1a,and reduced TXB2/6-K-PGF1a.There were little harm to gastric.It was very wholesome to the prognosis.TXB2 and 6-K-PGF1a should be the other detection indexes to AR.They were very useful to prediction and screening of AR,and they could be the references to the medical treatment in clinical.There were significant to judge and monitor the condition and to evaluate the prognosis. |
