ObjectiveTo investigate the correlation between the specific expression of mRNAs and AIDS in Heat-toxic Stagnation Syndrome,and to find the mRNA specific expression characteristics in AIDS syndromes,to explore the biological nature of AIDS Heat-toxic Stagnation Syndrome,and provide experimental evidence for the objective TCM syndrome diagnosis of AIDS.MethodsThis study carried out 10 mRNAs that significant specific expressed in Heat-toxin Stagnation Syndrome through access to relevant documents and analysis data,in the basis of the results of preliminary microarray.10 genes are:CHP1,APOBEC3F,CDA,GGT1,CHI3L1,ZEB1,KLF10,ATAD1,SP3,IL7R.Using real-time quantitative PCR(Quantitative Real-time PCR,qRT-PCR)for the acquisition of HIV Heat-toxin Stagnation Syndrome,Heat-toxin Stagnation Syndrome blood samples and normal control group,the above object of the mRNA expression of mRNA the amount of testing to GAPDH as a reference gene to standardize on 2-△ CT value of the target gene is expressed relative expression level.Use statistical analysis software SPSS V20.0 difference between the target mRNA of the AIDS group,asymptomatic group and normal control group,the expression of the relationship.Results1)The AIDS group and normal control group:CHI3L1,CDA,APOBEC3F downregulated in toxic hot Accumulation card of AIDS patients,and the difference was statistically significant(P<0.05),consistent with the microarray results;CHPI,GGT1,ZEB1,KLF10,ATAD1 isogenic although downregulation screening results are consistent with the chip,but the difference was not statistically significant(P>0.05).SP3,IL7R no significant difference,is also inconsistent with the chips down on the results of the expression.2)Asymptomatic group and normal control group:CHP1,APOBEC3F,CDA,GGT1 toxic hot Accumulation of AIDS syndrome patients downregulated,and the difference was statistically significant(P<0.05),consistent with the microarray results;CHI3L1,ZEB1,KLF10,ATAD1 isogenic although downregulation consistent with the microarray,but the difference was not statistically significant(P>0.05).SP3,IL7R no significant difference,is also inconsistent with the chips down on the results of the expression.2)AIDS on the group and asymptomatic group:CHI3L1 of AIDS and asymptomatic period were downregulated,and the difference between the two groups was statistically significant(P<0.05);CDA,APOBEC3F,CHP1,GGT1,ZEB1,KLF10,ATAD1,SP3,IL7R isogenic AIDS and asymptomatic period were downregulated,but there was no significant difference(P>0.05).Conclusions1.AIDS Heat-toxin Stagnation Syndrome and HIV Heat-toxin Stagnation Syndrome have differences in specific mRNA expression.AIDS Heat-toxin Stagnation Syndrome that specific mRNA expression is CHI3Ll,it’s mainly related to Thl/Th2 response,inflammatory cell activation and apoptosis,angiogenesis,tissue repair and remodeling and other functions,this may be the biological nature of AIDS Heat-toxin Stagnation Syndrome.HIV Heat-toxin Stagnation Syndrome that specific mRNA expression is CHP1、GGT1,it’s mainly related to growth and apoptosis of cells,regulation of inflammation,oxidative stress,inhibition of tumor cell growth and other functions,this may lead to the biological nature of HIV Heat-toxin Stagnation Syndrome.2.AIDS Heat-toxin Stagnation Syndrome and HIV Heat-toxin Stagnation Syndrome have a common specific mRNA expression:CDA、APOBEC3F.These two genes were no significant differences in the expression of these two periods,suggesting that these two genes and AIDS stage has nothing to do with HIV/AIDS Heat-toxin Stagnation Syndrome related.The main action of the genes involved in cytidine dehydrogenation reaction,inhibiting the replication of HIV,may play an important role in the pathophysiology of AIDS. |
