Objective To clarify the role of intervention of Jie Du Qu Yu Zi Shen Fang to TLR9-MyD88-NF-κB pathway.The research will provide the scientific basis of troditional Chinese medicine for the treatment of SLE.Methods Experimental study of the 30 MRL/lpr mices were randomly divided into model group,prednisone group,traditional Chinese medicine group.For each treatment group were appropriate drug theray for 8 weeks.By HE staining,PAS staining,Masson staining respectively MRL/lpr mouse kidney sections were stained pathology to observe and compare the pathological changes of three groups of mices.Using the method of Real-time PCR to compare the expression of differences of TLR-9,MyD88,NF-κBmRNA in each group of mice.Results HE staining of renal tissue showed,model control group presented with moderate to severe mesangial cells increased significantly widened mesangial matrix,showed segmental glomerular sclerosis,capillary endothelial cells proliferated,stenosis obvious prednisone kidney The most obvious improvement pathology;PAS staining showed,model group showed significant positive reaction,mesangial cells and matrix were significantly increased mesangial,renal tubular basement membrane and have seen a lot of lymphocyte infiltration,improve the prednisone group most significant renal pathology Masson staining,the model control group showed a lot of blue collagen deposition,showing segmental glomerular sclerosis,a large number of cell proliferation,glomerular lobulated renal interstitial fibrosis,glomerular capillary wall significant thickening,mesangial,basement shows a large red addicted complex deposition,the most obvious improvement in the prednisone group pathology.Real-time PCR assay showed that,the expression levels of TLR-9,MyD88,NF-κB mRNA in each treatment group were lower than in the model group TLR-9 mRNA expression levels,TCM group,prednisone group was significantly lower(P<0.05).The difference between TCM group and the prednisone group was not statistically significant(P>0.05).MyD88 mRNA expression levels,MyD88 mRNA expression levels of TCM group and prednisone group were significantly lower than those in the model(P<0.05)TCM group and prednisone group had no significant difference(P>0.05),NF-κB mRNA expression levels,the expression of NF-κB mRNA in TCM group and prednisone group was significantly lower than those in the untreated group(P<0.05),there was no significant difference in expression levels between traditional Chinese medicine group and prednisone group(P>0.05).Conclusion ① Jie Du Qu Yu Zi Shen Fang can protect the kidney of MRL/lpr mice and have an impact on its pathological changes,reducing kidney damage,delaying the course.②Jie Du Qu Yu Zi Shen Fang have an impact on TLR-9-MyD88-NF-KB signaling pathway of MRL/lpr mice,so as to achieve a protective role.for kidney.
