Objective:By establishing the models of anti-reflux esophagitis in rabbit induced by acid and bile,to research the treatment effect of Xuanfudaizhe decoction and groups for reflux esophag-itis.Using nifedipine,U-73122,2-APB,TG to block calcium channels,then comparing differen-ces in the magnitude of the tension of calcium channel blocked around among groups.And detecting the levels of VIP,SP,NOS2 in esophageal mucosa.Inquring into the regulatory role and mechanism of Xuanfu Daizhe decoction and groups for reflux esophagitis calci-um channel of LES deeply.Methods:Preparation the rabbit model by using HCL and bile(0.1 mol/LHCL 30ml and 5%bile 30ml,60min,ltime/day,for7days).Xuanfudaizhe decoction and groups were given soups c-orrespondingly after perfusing(10ml/kg).The model group and normal group were given the s-ame amount of saline(ltime/day,for10days).Restoring the LES and the lower esophageal.aft-er the rabbit died.Detecting the pathological changes of esophageal mucosa,using nifedipi-ne,U-73122,2-APB,TG to block calcium channels,then determining the LES muscle tension,using immunohistochemical method to detecte the levels of VIP,SP,NOS2.Results:The model group was lower than the normal group,the whole side group,gan liters group,the group went to the bitter drop,the group went to lifting and declining about calcium release and flow phase of LES(P<0.05 or P<0.01).There was significant difference between the model group and bitter drop group,the lifting and declining group,the group losing gan about calcium release phase of LES in ROCC(P<0.01).There was no significant difference b-etween the model group and bitter drop group,the lifting and declining group,the group losi-ng gan about calcium release phase of LES in L-type calcium channels and SERCA(P>0.0-5).There was no significant difference between the normal group and the whole side group a-bout calcium release and flow phase of LES(P>0.05).Compared with the model gro-up,there were significantly decreased about esophageal mucosa VIP expression in gan liters group(P<0.01).Compared with model group there was a significant difference about the expression of SP group(P<0.01)in the group went to lifting and declining.There were also a signi-ficant difference between the model group and the group went to the bitter drop(P<0.01).Conclusion:Xuanfu Daizhe decoction can keep cytosolic calcium ion dynamic equilibrium t-hrough opening L-type calcium channel activation ROCC,and regulation of SERCA.Gan lite-rs group had specific effect on L-type calcium channels.Xuanfu Daizhe decoction by reducing the expression of VIP and NOS2 and enhancing the expression of SP regulate the gastrointesti-nal motility to inhibit gastrointestinal contents reflux and promote mucosal repair.lt had best therapeutic effect.
