Objective:Background:It is generally believed that breast cancer is caused and induced by genetic and environmental factors jointly.However,except for the social cultures and lifestyle variations,reports on different genetic backgrounds between different ethnic groups were not thorough.Therefore,it is very essential to provide systematic research on the genetic factors between Uygur and Han patients with breast cancer.The purpose of this research is to detect the expression status of DNA repair genes,BRCA1,BARD1 and XRCC1 in breast carcinoma tissue of Uygur,Han patients,and analyze the relationship with the occurrence of breast cancer and racial differences.Methods:70 breast cancer patients of Uygur and Han ethnic groups were chosen,and real-time fluorescence quantitative PCR determination was applied to detect the mRNA expression of BRCA1、BARD1、XRCC1 in their tissue.We randomly selected 54 cases,27 in each group,and detected the expressions of BRCA1,BARD1 and XRCCIProtein with Western blot method.Results:mRNA and protein expression of BRCA1 in both Uygur,Han breast carcinoma tissue were lower than the corresponding adjacent tissues(P<0.05),and there were statistically significant differences between two ethnic groups(P<0.05).mRNA and protein expression of BARD1 in the both groups were lower than the corresponding adjacent tissues(P<0.05),with no statistically significant differences between two groups(P>0.05).mRNA expression of XRCC1 of carcinoma tissue was higher than the corresponding adjacent tissues(P<0.05),while theprotein expression was lower(P<0.05),and there were statistically significance in transcription levels(P<0.05).Conclusion:there are different transcription and translation expressions in BRCA1,BARD1,XRCC1 between breast cancer tissues and normal tissues,and racial differences arefound. |
