Objective: To explore human micro RNA let-7a1,let-7c and CDC25 A gene expression in lung cancer tissues,para-carcinoma tissue and Benign lung disease tissues and with the relationship between the expression and clinical pathological features.Method:Selected and collected the 53 cases of surgical specimens of lung tissue,Including 44 cases of lung cancer tissue,respectively,to collect carcinoma tissues and normal tissue adjacent to carcinoma tissues(pathological confirmed);9 cases of benign lung disease group.and normal tissue adjacent to carcinoma tissues,and 9cases of benign pulmonary disease tissues.Trizol method was used to extract total RNA,reverse transcription by adding tail method fluorescence quantitative PCR detection of gene expression.Immunohistochemical method to detect CDC25 A protein expression.Results:Let-7a1 and let-7c gene expression in lung cancer tissue(0.93±0.46,1.16±0.76)significantly lower than the normal tissue adjacent to carcinoma(1.18±0.54,1.59±0.88)and Benign lung disease tissues(1.38±0.54,1.81±0.63),statistically significant difference(F=4.397,4.189,P<0.05),5 cases of late clinical stage(Ⅲ-Ⅳperiod)the expression of let-7a1(0.50±0.28)significantly lower than the early(Ⅰ-Ⅱperiod)the expression of let-7a1(0.99±0.45),the difference was statistically significant(P < 0.05);Let-7c expression decline associated with lymph node metastasis.Let-7a1 and let-7c genes are expressed in terms of gender differences of no statistical difference(P > 0.05).Let-7a1 in lung cancer differentiation of malignant degree is more obvious,and the let-7c express only significantly decreased between the patients with malignant degree is higher and lower.At the same time,the study found that the let-7a1 gene expression was significantly negative correlation with smoking,and expression of let-7a1 gene smoking patients(0.63±0.21)significantly lower than nonsmoking patients(1.27±0.41),significant difference(P < 0.05),with statistical significance.CDC25 A protein in the cancer tissue,the tissue adjacent to carcinoma and benign pulmonary disease tissue is expression of obvious differences(χ2=33.351,P<0.05).CDC25 A expression in lung cancer group is significantly higher than the latter two(P<0.05).CDC25 A protein expression in lung cancer tissue have nothing to do with age,gender,pathological type,degree of tumor differentiation,clinical stages(P > 0.05),and Associated with smoking,lymph node metastasis(P <0.05).CDC25 A gene m RNA expression was significantly higher in lung cancer tissue than the tissue adjacent to carcinoma and benign pulmonary disease(F=6.333,P<0.05).Pearson correlation analysis shows that between CDC25 A and let-7c gene expression in lung cancer tissues and adjacent tissues were significantly negative correlation(r Carcinoma tissues=-0.423,r para-carcinoma tissue=-0.397,P < 0.05),the difference was statistically significant.Conclusion:(1)the let-7 a1,let-7 c and CDC25 A can be used as one of the molecular markers to diagnose lung cancer;And may become a new target for lung cancer gene therapy;(2)the let-7 c lower expression and CDC25 A high expression have promote lung cancer lymph node metastasis.(3)CDC25A is probably one of the 7 c downstream target genes;(4)the let-7 a1,let-7 c expression changes may have decided to lung cancer differentiation respectively;(5)smoking may cause CDC25 A and let-7 a1 expression change,but there is no correlation between the two,has provided the new mentality for tobacco induced lung cancer development.
