Objective To analyse the effects of reveratrol on body weight and reproductive system of Sprague-Dawley rats with high fat diet.Methods Male and female Sprague-Dawley rats at the age of three weeks old(n=48)fed with standard diet one week for adapatability,then random blocks design besed on their weight was used to divide different gender rats into four groups respectivly:standard diet control group(SC,standard diet),high fat control group(HFC,high fat diet),Resl group(high fat diet with resveratol 6mg/kg·d-1),Res2 group(high fat diet with resveratrol 18 mg/kg·d-1).Rats were housed six per cage.Resveratrol was given through an orogastric catheter,SC and HFC group were given the same volume saline as resveratrol groups.Body weight and the naso-anal lenth measured indicator of linear growth for lee index were evaluated once a week while vaginal smear was used to examine the oestrous cycle.All rats were sacrificed after elven weeks of intervention by took,blood from arteia crualis,perirenal fat was collected and quickly frozen in liquid nitrogen and stroed at-80 ℃ until further processing.Levels of plasma estrodol and testerone were measured by radioimmunoassay.Commercial kits for Real Time PCR and enzyme-linked immunosorbent assay(ELISA)were used to measure Sirtl,LKB1,AMPK,ACC mRNA and content from adipose tissue.Pathdogical alterations of testis,epididymis,uterus,ovary were observed using optics microscope.Results 1.Compared with SC group,the gian of weight in HFC group was faster and lee index was markly higher(P<0.05,P<0.01)while resveratrol could restrain the high fat dietary induced high speed of weight increase,there was no significant differences in body weight,among SC group,Reslgroup and Res2 group.2.HFC group had a significantly lower level of Sirtl,LKB1,AMPK and higher level of ACC compared to SC group(P<0.01).Meanwhile,resveratrol could resverse this situaion.Resveratrol could enhanced the expression levels of Sirtl,LKB1 and AMPK and decreased the expressiong of ACC compared with HFC group(P<0.05,P<0.01),though they got markly improved than HFC group still had significant differences compared with SC group(P<0.05).3.The plasm testerone was markly lower in male HFC group(P<0.01),treatmenting with resveratrol,plasm testerone were markly enhanced(P<0.01)and had no significant differences compared with SC group.Testis coefficient was obviously lower in male rats HFC group than that in SC group(P<0.01).Uterine coefficient was distinctly higher in female HFC group compared to SC group(P<0.01).The result of HE satining showed that some of seminiferous tubules and epididymal lumen were atrophic in male rats HFC group.Spermatogeinc cells ablated and cumulated in lumen and the mature sperms reduced in epididymal lumen under high power leans.Proliferation of smooth muscle of uterine and anomalism of ovarian follicle were observed in famale HFC group.Parts of cells had lipoid degeneration under high power leans.Reproductive organs were regular and integral and there was less lipoid degeneration cells in Res 1 and Res2 group.Conclusion 1.Resvertral could prevent the high fat diet induced body weight increase in weaning rats until sexual maturity.The mechanism may partly because resveratrol could upregulate the level of Sirtl,LKB1,AMPK and downregulate ACC in adipose tissue.2.Resveratrol prevent high fat diet induced impairment in reproductive system,we did not observed the effects of resvertarol in reproductive system during administration.
