PDCH10 Promotes Apotosis In Endometrioid Endometrial Adenocarinoma Cell

Objective: To investigate the apoptosis-inducing effect of protocadherin 10 gene(PCDH10)on endometrioid endometrial adenocarinoma(EEC)cells and the underlying mechanisms.Methods:Both human EEC cell lines,AN3 CA and HEC-1-B,were transiently transfected with PCDH10 expression plasmid or control Vector by lipofectamine 2000.The expression of PCDH10 at mRNA and protein levels in the PCDH10-transfected cells were detected by real-time PCR and Western blotting respectively.Cell apoptotic rate was examined by Tunel Assay;The expression of Apoptosis related protein were measured by Western blotting.Results: PCDH10 could be successfully transfected into AN3 CA and HEC-1-B cell lines by transient transfection using lipofectamine 2000,Tunel assay revealed that ectopic expression of PCDH10 in EEC cell may induce apoptosis.Mechanistic studies revealed that PCDH10 induced EEC cell apoptosis by down-regulating Bcl-2,and up-regulating Fas 、caspase3、caspase8 and caspase9 factors.Conclusions: PCDH10 could significant promote tumor cell appotosis via caspase-dependant pathway,which may be used as potential therapeutic targets.
