The Effect On Characterization Of Aging And Possible Mechanism Of Traditional Chinese Herb Polygonum Multiflorum Extractive Tetrahydroxystilbene Glucoside

Aging is an inevitable process of life caused by a combination of organs and tissues events which manifests as loss of structure and function.Aging is accompanied by hypofunction irritability,energy metabolism lessen,abnormal cell structure and mitochondria reduction,leading to death.Therefore,It is critical to do research about the mechanism of aging,as equal to find new drugs to slow aging and death.Polygonum multiflorum as a traditional Chinese medicine has been widely used to anti-aging.Doctors of traditional Chinese medicine speak highly of its anti-aging effect.Interestingly,its extractive has been proved to prolong lifespan in fruit fly.2,3,5,4'-tetrahydroxystilbene-2-o-b-d-glucoside(TSG)as the main ingredient of Polygonum multiflorum,has similar structure with resveratrol which shows great anti-aging effect.However,the mechanism of TSG to anti-aging is still unknown.In our study,we did research about how TSG slow aging with high-fat induced model by regulating aging-related pathway.The result showed that TSG and resveratrol could increase the percentage of active AMPK without changing the content of SIRT1.To further study on SIRT1 pathway,we detected the mRNA alternation of downstream of SIRTI pathway,such as PGC-Ia,NRF-1,TFAM,CytC,HSP60,ClpP and NAMPT by RT-PCR.The result reflected that both of TSG and resveratrol can increase the expression of NAMPT to stimulate SIRT1 activity.And it could also increase the mRNA level PGC-la,NRF-1,TFAM,CytC at the same time.However,it did no effect on HSP60 and ClpP.At last,we used high content screening to dectect the alternative of mitochondrial mass.And we finally discovered that TSG in high concentration has the same effect of increasing mitochondrial as resveratrol.Animal study was used to evaluate the effect of life extension of TSG.We used high calorie food to accelerate aging,and were randomly assigned the mice into two groups:TSG and High Calorie food,compared with control group.We found that TSG has a tendency of prolong mean lifespan,and improved motor ability and bone density.
