Effect Of 2, 3, 5, 4′-tetrahydroxystilbene-2-o-β-d-Glucoside On The Expression Of NF-κB/IκB Of Huvec Injured By H₂O₂

Aim: In this report, we observed the cytoprotection effect and molecular mechanism of 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside on isolated human umbilical vein endothelial cell injured by H₂O₂.Methods: In the studies, human umbilical vein endothelial cells were treated with H₂O₂ and TSG in vitro. Cell growth and cell viability were determined by MTT assay. The HUVEC was treated with 200μmol/L H₂O₂ for 24h after pretreated with different concentrations of TSG for 4h.Whereafter, the mRNA levels of NF-κB, IκB, TNF-αand IL-6 in the HUVEC were determined by RT-PCR. Western-blot analysis was used to detect protein levels of NF-κB and IκB. Protein levels of TNF-αand IL-6 in the supernatants of cells were determined by ELISA.Results: The injured concentration of 200μmol/L H₂O₂ was selected by MTT. The cell proliferation rate evaluated by MTT showed that 0.1, 1, 10μmol/L TSG plus H₂O₂ significantly increased (P < 0.01)the cell proliferation rate compared with H₂O₂ (200μmol/L) group. Compared with H₂O₂(200μmol/L) group, the expression of NF-κB mRNA all had significant decreased(P<0.01) , but there was only group 1μmol/L TSG plus H₂O₂ had the significant difference in the expression of NF-κB protein. The level of TNF-αmRNA and protein were reduced. The level of TNF-αmRNA decreased significantly (P<0.01) in 1μmol/L TSG plus H₂O₂ group. The expression of IκB and IL-6 had no significant differences among all groups.Conclusion: TSG might reduced the expression of inflammatory factors via inhibiting NF-κB activion and protected HUVEC.
